Improvment of the Quality Standard of Jianpiyiqi Mixture

摘要 目的完善健脾益气合剂的质量标准。方法采用薄层色谱法定性鉴别黄芪、制何首乌、白术和陈皮4味药材。采用高效液相色谱法测定陈皮主要成分橙皮苷的含量,色谱柱为WondaCract ODS-2柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(20∶80,V/V),流速为1.0 m L/min,进样量为10μL,柱温为30℃,检测波长为285 nm。结果黄芪、制何首乌、白术和陈皮的图谱清晰,与杂质斑点分离较好,阴性对照无干扰;橙皮苷质量浓度在7.6~121.6μg/m L(r=1.0000)范围内与峰面积线性关系良好,平均加样回收率为100.99%,RSD为1.45%(n=8)。结论该方法简便、重复性好、专属性强,可用于健脾益气合剂的质量控制。 Objective To improve the quality standard of Jianpiyiqi Mixture.Methods Thin layer chromatography(TLC)was used to qualitatively identify the four herbs including Astragali Radix,Polygoni Multiflori Radix Preparata,Atractylodis Macrocephalae Rhizoma,and dried orange peel.High performance liquid chromatography(HPLC)was used for the quantitative analysis of the main component of hesperidin in dried orange peel.The chromatographic column was a WondaCract ODS-2 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid solution(20∶80,V/V),and the flow rate was 1.0 m L/min,the injection volume was 10μL,the column temperature was 30℃,and the detection wavelength was 285 nm.Results The TLC spots of Astragali Radix,Polygoni Multiflori Radix Preparata,Atractylodis Macrocephalae Rhizoma,and dried orange peel were clear;the spots were well separated and the negative control had no interference.Hesperidin indicated good linearity in the range of 7.6-121.6μg/m L with the peak area(r=1.0000);the average recovery rate(n=8)were 100.99%,and RSD was 1.45%(n=8).Conclusion Thiss method is easy,reproducible,and specific,which can be used as the quality standard of Jianpiyiqi mixture.
