摘要
目的检测长链非编码RNA(LncRNA)小核仁RNA宿主基因7(SNHG7)在食管鳞癌(ESCC)组织中的表达及与患者临床病理特征和预后的关系。方法选取2014年1月至2015年1月间陕西省核工业215医院收集的89例ESCC患者的术后肿瘤组织和对应癌旁组织标本,采用实时荧光定量聚合酶链式反应(PCR)法(qRT-PCR)检测SNHG7表达情况,分析ESCC组织中SNHG7的相对表达量及与患者临床病理特征和预后的关系。结果SNHG7在ESCC肿瘤组织中的表达水平为(4.15±0.83),在癌旁组织中为(1.04±0.17),差异有统计学意义(P<0.05)。以SNHG7在ESCC肿瘤组织中表达水平的平均数为界值,将患者分为SNHG7低表达组和高表达组。SNHG7表达与ESCC患者性别、年龄、病变位置和浸润程度无关,差异无统计学意义(P>0.05),与TNM分期、有无淋巴结转移和分化程度有关,差异有统计学意义(P<0.05)。SNHG7高表达ESCC患者和SNHG7低表达患者3年生存率分别为39.6%和77.8%,差异有统计学意义(P<0.05)。单因素分析显示,SNHG7表达、肿瘤浸润程度、TNM分期、有无淋巴结转移和肿瘤分化程度均是影响ESCC患者预后的危险因素,差异均有统计学意义(P<0.05)。多因素分析显示,SNHG7表达和肿瘤分化程度是影响ESCC患者预后的独立危险因素,差异均有统计学意义(均P<0.05)。结论SNHG7在ESCC组织中表达水平高,SNHG7的高表达与食管鳞癌患者的临床分期、淋巴结转移、肿瘤分化及预后差有关。
Objective To detect the expressions of long non-coding RNA(lncRNA)small nucleolar RNA host gene 7(SNHG7)in esophageal squamous cell carcinoma(ESCC),and to investigate the relationships between the expression and clinicopathological features and prognosis.Methods Tumor tissues and corresponding paracancerous tissues were collected in 89 ESCC patients.The expression of SNHG7 was detected by quantitative real-time fluorescence quantitative PCR(qRT-PCR),and the relationship between the expression of SNHG7 in ESCC tissues and clinicopathological characteristics and prognosis was analyzed.Results The expression level of SNHG7 was 4.15±0.83 in ESCC tissues and 1.04±0.17 in adjacent tissues(P<0.05).The expression of SNHG7 in ESCC was considered as cut off value and the patients were divided in a high expression group and a low expression group.The analysis showed that the expression of SNHG7 was associated with TNM stage,lymph node metastasis and tumor differentiation(all P<0.05)rather than with gender,age,lesion location and infiltration degree(P>0.05).The 3-year survival rate was 39.6%and 77.8%,respectively in patients with high expression of SNHG7 and patients with low expression of SNHG7(P<0.05).Univariate analysis showed that the expression of SNHG7,degree of tumor invasion,TNM stage,lymph node metastasis and and tumor differentiation were risk factors influencing the prognosis of ESCC patients.Multivariate analysis showed that expression of SNHG7 and tumor differentiation were independent risk factors for prognosis of ESCC patients(all P<0.05).Conclusion The expression of SNHG7 is elevated in ESCC tissues,and the high expression of SNHG7 is associated with the clinical stage,lymph node metastasis,tumor differentiation and poor prognosis of ESCC.
