聚乙烯醇-羧甲基壳聚糖复合水凝胶药物缓释体系的制备及评价 被引量:13

Preparation and evaluation of PVA-CMCS composite hydrogel for sustained drug delivery system

摘要 优化设计的功能水凝胶是一种优良的药物缓释载体。以聚乙烯醇(PVA)和羧甲基壳聚糖(CMCS)为原料,以两种物理交联方法相结合,制备了具有互穿网络结构的PVA-CMCS复合水凝胶。通过形貌特征分析、X射线分析、热重分析、红外分析和溶胀性能等一系列表征考察了复合凝胶的物理化学性能。探索了PVA-CMCS复合水凝胶的最佳制备条件,并进行了载药和释药行为研究。结果发现:CMCS浓度、甘油磷酸钠(GP)用量及冻融次数均会影响复合水凝胶的形态和溶胀性能。其中5%PVA与3%CMCS等量混合、加入0.6g GP、冻融4次的复合水凝胶,在形态和溶胀上的结果最好。然后分别以大分子药物牛血清白蛋白(BSA)和小分子药物盐酸二甲双胍(MH)为模型药物,经释药研究表明,该复合水凝胶具有明显的pH敏感性,而且对大分子药物表现出更好的缓释效果。该复合凝胶制备方法简单温和,在作为大分子药物缓控释输送载体方面具有潜在的应用前景。 The optimized functional hydrogel is an excellent sustained-release drug carrier.Polyvinyl alcohol(PVA)/carboxymethyl chitosan(CMCS)composite hydrogels with interpenetrating network were prepared.A series of characterizations,such as X-ray analysis,thermogravimetric analysis,infrared analysis and swelling property,were carried out.The optimal performance conditions of the hydrogel were investigated,and the drug release experiment was carried out by preparing PVA-CMCS loaded hydrogel.The results showed that the morphology and swelling properties of the hydrogels were affected by the concentration of CMCS,the dosage of sodium glycerophosphate(GP)and the number of freeze-thaw times in PVA-CMCS hydrogels.If the optimal preparation condition was that the volume of 5%PVA was equal mixing to the volume of 3%CMCS and 0.6g GP,as well as freeze-thawed for 4times,the hydrogels had the best results in morphology and swelling behavior.Under these conditions,the drug-loaded hydrogels containing macromolecular bovine serum albumin and small metformin hydrochloride were prepared respectively.The results of drug release experiments showed that PVA-CMCS exhibited notably pH sensitivity as well as good performance in sustained release of macromolecular drugs.Furthermore,PVA-CMCS composite hydrogels prepared by the simple and mild process had great potential to be used as sustained drug delivery carriers for macromolecular drugs.
