miR-125b靶向调节p53表达对生精细胞发育能力的影响

摘要 目的:分析miR-125b靶向调节p53表达对生精细胞发育能力的影响。方法:选取出生7d的清洁级健康小鼠,以颈椎脱臼法将其处死,取出睾丸,进行支持细胞培养,以miR-125b的inhibitor与mimic对小鼠支持细胞进行转染,采取四甲基偶氮唑盐法对细胞的增殖情况进行检测,并测定增殖细胞核抗原表达及p53表达。结果:miR-125b可使p53表达下调,p53表达是miR-125b的靶基因。将p53的小干扰RNA转染支持细胞后,发现p53基因沉默可对支持细胞组织增殖产生促进作用。结论:miR-125b通过靶向调节p53表达,从而促进生精细胞发育能力的提升。 Objective:To analyze the effects of miR-125b on the development of spermatogenic cells by targeting the regulation of p53 expression.Methods:7days clean healthy mice were selected,sacrificed by cervical dislocation,testicles were removed,and the supporting cells were cultured.The supporting cells were transfected with inhibitor and mimic of miR-125b.The proliferation of the cells was detected by tetramethylazolium salt method,and the expression of proliferating cell nuclear antigen and p53 were measured.Results:miR-125b down-regulated the expression of p53,which was the target gene of miR-125b.After transfection of p53 small interfering RNA into supporting cells,it was found that p53 gene silencing could promote the proliferation of supporting cells.Conclusion:miR-125b promotes the development of spermatogenic cells by targeting the regulation of p53 expression.
