向日葵WRKY转录因子家族鉴定与生物信息学分析 被引量:5

Genome-wide Identification and Bioinformatics Analysis of WRKY Transcription Factor Family in Sunflower(Helianthus annuus)

摘要 WRKY转录因子家族参与了多种激素调节信号通路,在帮助植物响应外界不良环境中起着重要作用。本研究基于向日葵(Helianthus annuus)基因组数据库对向日葵进行了WRKY基因家族成员鉴定、进化关系分析、基因定位、基因结构和保守Motif分析、功能启动子元件和基因共线性分析。结果表明:向日葵中包含117个WRKY家族基因,分为Ⅰ、Ⅱ、Ⅲ类,其中Ⅱ类包含Ⅱa、Ⅱb、Ⅱc、Ⅱd和Ⅱe五个亚家族。染色体定位显示HaWRKY基因在向日葵所有染色体上均有分布,第10号染色体上分布最多含有18个HaWRKY基因,2号和13号染色体分布最少仅有3个HaWRKY基因。向日葵WRKY转录因子蛋白在59~940个氨基酸之间,平均氨基酸个数为345个,等电点为5.01~10.19不等。基因结构分析表明除Ha WRKY52、HaWRKY82不含内含子,其他成员都含有1~9个不等的内含子。保守域分析表明有12个HaWRKY家族基因WRKYGQK发生了变异,占向日葵WRKY家族的10%。此外,启动子分析表明HaWRKY家族基因启动子区含有大量响应生物胁迫和非生物胁迫元件,共线性分析得到11对加倍复制基因。本研究结果有助于了解向日葵WRKY基因家族成员的情况,为向日葵WRKY基因家族成员进一步的功能分析提供基础。 The WRKY transcription factor family is involved in various hormone regulatory signaling pathways and plays an important role in helping plants respond to adverse external environments. Based on the Helianthus annuus genome database, we identified the members of the WRKY gene family and analyzed evolutionary relations, gene locations, gene structures, conserved motifs, functional promoter elements and gene collinearity. The results showed that 117 HaWRKY genes were identified there were 117 WRKY family genes in sunflower, which could be classified into three main groups(Ⅰ, Ⅱ, and Ⅲ), and GroupⅡ could further be divided into five subgroups(Ⅱa, Ⅱb, Ⅱc, Ⅱd, and Ⅱe). Chromosomal location display HaWRKY genes distributed on sunflower all chromosomes. There are 18 HaWRKY genes on chromosome 10, which is the largest number of WRKY genes. At least three HaWRKY genes were found on chromosomes 2 and 13. HaWRKY proteins contained 59 to 940 amino acids,with 345 amino acids in average and the isoelectric point ranged from 5.01 to 10.19. Gene structure analysis showed that only HaWRKY52 and HaWRKY82 had no intron, other members all contained 1-9 introns. Conserved domain analysis showed that 12 genes of the Ha WRKY family WRKYGQK were mutated, accounting for 10% of the sunflower WRKY family. In addition, promoter analysis showed that the promoter region of the HaWRKY family gene contained a large number of elements in response to biological and abiotic stress. And 11 pairs of double copy genes were obtained by collinearity analysis. The results of this study are helpful to understand the situation of sunflower WRKY gene family members and provide a basis for further functional analysis of sunflower WRKY gene family members.
