摘要
甘露糖结合凝集素(mannose binding lectin,MBL)是一种钙离子依赖的糖结合蛋白,是天然免疫的重要组成部分。本研究旨在探究牦牛MBL2基因mRNA组织表达谱及可能靶定它的miRNAs组织表达谱。本研究首先应用RT-PCR技术分析牦牛MBL2基因mRNA在牦牛肝、脾、肺、肾、后腿骨骼肌、卵巢、小肠、颌下淋巴结、大肠、肠系膜淋巴结这10种组织里的表达谱,其次使用Targetscan和miRBase软件针对可能靶定MBL2基因的miRNAs进行预测,最后利用RT-PCR技术分析可能靶定MBL2基因的miRNAs在牦牛所检测10种组织中的表达谱。试验结果显示,MBL2基因mRNA在所检测的牦牛组织中仅在肝中表达,表达水平极显著高于脾、肺、肾、后腿骨骼肌、卵巢、小肠、颌下淋巴结、大肠、肠系膜淋巴结组织(P<0.01);预测到可能靶定牦牛MBL2基因的miRNAs共有37个,从中选择了8个进行组织表达谱分析,发现bta-miR-143、bta-miR-193a、bta-miR-345-5p、bta-miR-2385-3p、bta-miR-2432、bta-miR-2285p、bta-miR-2284t-5p、bta-miR-126-5p在各组织中均有表达,与MBL2基因的mRNA在牦牛肝中共表达。综上所述,牦牛MBL2基因可能主要由肝表达,预测筛选的8个miRNAs可能在肝中通过靶定MBL2基因发挥调节作用,但它们之间是否存在真实的靶向关系及其靶向调节的生物学效应都有待于进一步深入研究。
Mannose binding lectin(MBL)is a calcium-dependent glycoconjugate protein that is an important component of natural immunity.The aim of this study was to investigate the mRNA tissue expression profile of yak MBL2 gene and its possible targeted miRNA expression profile.This study firstly analyzed the mRNA expression profile of yak MBL2 gene in ten tissues of liver,spleen,lung,kidney,muscle,ovary,small intestine,submandibular lymph node,large intestine and mesenteric lymph node by RT-PCR.Targetscan and Micro Base software were used to predict the miRNAs that might target MBL2 and RT-PCR was used to analyze expression profiles of miRNAs that might target MBL2 in the ten yak detected tissues.The results showed that MBL2 gene mRNA was only expressed in the yak liver,and the expression level was significantly higher than that of spleen,lung,kidney,muscle,ovary,small intestine,submandibular lymph node,large intestine and mesenteric lymph node(P<0.01).A total of thirty-seven miRNAs were predicted to target yak MBL2 gene,and eight of them were selected for tissue expression profiling.The results showed that bta-miR-143,bta-miR-193 a,bta-miR-345-5 p,bta-miR-2385-3 p,bta-miR-2432,bta-miR-2285 p,bta-miR-2284 t-5 p and bta-miR-126-5 p were expressed in all detected tissues and co-expressed with MBL2 mRNA in yak liver.In summary,the yak MBL2 gene is mainly expressed by the liver,and the eight predicted miRNAs may play a regulatory role in the liver by targeting MBL2 gene,but whether there is a real targeting relationship between them and the biological effects of targeting regulation need to be further studied.
