摘要
为建立家禽样本中的禽白血病病毒抗原化学发光免疫分析检测方法,本研究通过获得的禽白血病病毒P27融合蛋白与相关单克隆抗体建立了家禽样本中的禽白血病病毒抗原化学发光检测试剂盒。该方法能够特异性地定量检测家禽样本中的禽白血病病毒抗原,敏感性高,特异性强,对其他相关的禽类病原无交叉反应。对该方法的特异性指标、敏感性指标进行了验证,同进口的禽白血病病毒抗原ELISA检测试剂盒进行了比较,结果显示,阳性样品符合率为92.63%,阴性样品符合率为93.17%,总符合率为93.98%。重复性试验的组内与组间变异系数均小于10%,具有良好的重复性。上述结果表明,本研究建立的禽白血病病毒抗原化学发光免疫分析检测方法特异性高、敏感性强,操作简单、快速,具有较高的应用推广价值。
To establish a chemiluminescence immunoassay method for the detection of avian leukemia virus antigen in poultry samples,a chemiluminescence assay kit for avian leukemia virus antigen in poultry samples was established through the fusion protein of avian leukemia virus P27 and related monoclonal antibodies.This method could quantitatively detect avian leukemia virus antigen in poultry samples,with high sensitivity and specificity,and no cross-reaction to other related avian pathogens.The specificity and sensitivity of this method were verified.Compared with imported avian leukemia virus antigen ELISA kit,the coincidence rate of the positive samples was 92.63%,the coincidence rate of the negative samples was 93.17%,and the total coincidence rate was 93.98%.The coefficient of variation within and between groups was less than 10%,showing good repeatability.The chemiluminescence immunoassay method of avian leukemia virus antigen established in this study has high specificity,high sensitivity,simple and rapid operation,and has high application and promotion value.
