响应面法优化纤维素酶辅助提取蛹虫草中虫草素工艺 被引量:4

Optimization of Cellulase-Assisted Extraction of Cordycepin in Cordyceps militaris by Response Surface Method

摘要 响应面法优化纤维素酶辅助提取人工子实体蛹虫草(Cordyceps militaris)中的虫草素,通过单因素试验,考察提取时间、酶添加量、pH和提取温度对虫草素含量的影响,在此基础上,应用二阶响应曲面的Box-Behnken设计优化虫草素的提取工艺。运用超高效液相色谱法(UPLC)对虫草素进行含量检测,确定色谱条件为:流动相甲醇-水(15︰85)、检测波长260 nm、流速0.5 mL/min、进样量10μL。结果表明,人工培养的蛹虫草子实体中虫草素热水浸提的最佳工艺条件为:提取时间120 min、酶添加量2.5%、pH 4.5、提取温度40℃,在这些条件下进行3次重复试验,测得蛹虫草中虫草素提取率为0.1726%。与回归方程预测值相近,表明纤维素酶辅助提取蛹虫草中虫草素的提取方法可行,为虫草素的提取提供研究基础和参考价值。 The effect of extraction time,enzyme addition,pH and extraction temperature on the content of cordycepin was optimized by using the response surface method to extract the cordycepin from the artificial solid Cordyceps militaris by single factor experiment,and on this basis,the extraction technology of cordycepin was optimized by using the Box-Behnken design of second-order response surface.The content of cordycepin was detected by ultra-high performance liquid chromatography(UPLC),and the chromatographic conditions were determined as follows:flow phase of methanol-water(15︰85),detection wavelength 260 nm,flow rate 0.5 mL/min,and injection volume 10μL.The results showed that the optimum technological condition of hot water extraction of cordycepin from cultured Cordyceps militaris fruiting bodies were extraction time 120 min,the amount of enzyme added 2.5%,pH 4.5,the extraction temperature 40℃,and 3 repeated experiments were carried out under this condition,and the extraction rate of cordycepin in Cordyceps militaris was 0.1726%.Similar to the predicted value of regression equation,the method of cellulase-assisted extraction of cordycepin from Cordyceps militaris was feasible,which provided further research basis and value for cordycepin extraction.
