摘要
以玫瑰茄超微粉为材料,通过单因素结合响应面分析,旨在对玫瑰茄中花色苷的乙醇浸提工艺进行优化。比较了乙醇体积分数、料液比、浸提时间、浸提温度等单因素对玫瑰茄花色苷提取率的影响。在单因素试验的基础上,根据中心组合(Box-Behnken)实验设计原理,采用三因素三水平的响应面分析法,以花色苷含量为响应值做响应面并进行回归分析。结果表明:玫瑰茄花萼超微粉总花色苷提取条件为提取剂为体积分数70%乙醇,提取温度为54℃,料液比为1︰21(g/mL),提取时间为101 min,测定总花色苷的含量为729.68 mg/100 g,比普通采用玫瑰茄花萼为原料提取所得的总花色苷含量要高得多。此次试验为玫瑰茄花萼超微粉在食品工业上的应用提供了一定的理论基础。
The conditions of extraction method were developed for the extraction of anthocyanins from superfine grinding roselle calyces powder based on response surface methodology(RSM).The single experimental factors,such as solvent concentration,solid-liquid ratio,extraction time,and extraction temperature,were studied.Experimental factors and their levels were determined by single factor tests.Subsequently,the Box-Behnken experimental design with three factors and three levels was performed,and the factors influencing the extracted technology were estimated by means of regression analysis.Response surface and contour were finally graphed with the extraction yield of anthocyanins as the response value.The optimized extraction parameters were obtained as follows:70%ethanol,1︰21(g/mL)for the solid-liquid ratio,101 min for the extracting time,and 54℃for the extracting temperature.Under this optimal condition,the yield of anthocyanins was 729.68 mg/100 g,which was quite higher than the similar extraction studies using roselle calyces.This experiment provided a theoretical basis for the application of roselle calyx ultrafine powder in food industry.
