TCGA数据库372例胃癌组织CXCL10基因表达及临床意义 被引量:2

CXCL10 gene expression and clinical significance of 372 cases of gastric cancer in TCGA database

摘要 目的胃癌是导致癌症死亡的主要原因之一,是东亚部分地区最常见的胃肠道恶性肿瘤。随着医疗水平的提升,胃癌患者5年生存率有所提高,但至今仍无法做到痊愈根治。本研究对癌症和肿瘤基因图谱(Cancer Genome Atlas,TCGA)数据库中胃癌和癌旁组织的基因测序数据进行挖掘,寻找胃癌治疗的新靶点。方法从TCGA数据库中获取372例胃癌和32例癌旁组织的RNA测序数据,按照logFC>1且P<0.05的标准筛选差异表达基因,通过WGCNA分析并关联胃癌患者的临床表型数据找出标志性基因。结果通过edgeR包和DESeq2包数据分析得到1 355个上调基因和1 774个下调基因。WGCNA分析差异基因得出11个模块,分别与患者临床数据进行相关性分析发现"black"模块与患者的生命状态呈正相关。将"black"模块中的基因进行蛋白质互作分析得到CXCL10基因节点度最高,经生存分析得出CXCL10基因与总生存率存在相关性,P<0.01。结论 CXCL10基因可能在胃癌的早期诊断和临床治疗中有重要的意义。 OBJECTIVE Gastric cancer is one of the leading causes of cancer death.It is the most common gastrointestinal malignant tumor in some parts of East Asia.With the improvement of medical standards,the 5-year survival rate has improved,but it still can not be cured.The purpose of this study was by mining the genetic sequencing data of gastric cancer and adjacent tissues in the TCGA database to find new targets for gastric cancer treatment.METHODS The RNA sequencing data of 372 cases of gastric cancer and 32 cases of adjacent cancer tissues were obtained from the TCGA database.Differentially expressed genes were selected according to the criteria of logFC>1 and P<0.05.The WGCNA analysis was used to correlate the clinical phenotypic data of gastric cancer patients and to find the signature genes.RESULTS According to the data analysis of edgeR package and DESeq2 package,1 355 up-regulated genes and 1 774 down-regulated genes were obtained.WGCNA analysis of differential genes yielded 11 modules that were correlated with patient clinical data and found that the " black" module was positively correlated with the patient’s life status.The interaction analysis of the genes in the " black" module showed that the CXCL10 gene had the highest node degree.After survival analysis,it was found that there was a significant correlation between the CXCL10 gene and the overall survival rate(P<0.01).CONCLUSION The CXCL10 gene may have important significance in the early diagnosis and clinical treatment of gastric cancer.
