摘要
目的多指标优选蒲公英的提取与纯化工艺。方法以咖啡酸、总酚酸、总黄酮为评价指标,通过正交试验确定蒲公英提取工艺;以咖啡酸、总酚酸、总黄酮转移率和浸出物收率为评价指标,通过正交试验确定纯化工艺。结果提取工艺为第一次加水12倍量,提取1.5 h,第二次加水10倍量,提取1.0 h,第三次加水10倍量,提取1.0 h。纯化工艺为浓缩密度1.05,醇沉浓度70%,静置12 h。结论蒲公英提取与纯化工艺稳定,合理可行。
Objective To optimize the extraction and purification process of Taraxaci Herba by multi-index evaluation method.Methods The extraction process is determined by orthogonal tests with caffeic acid,total phenolic acid and total flavonoid as the evaluation indexes.The purification process is carried out by orthogonal tests with the transfer rate and extraction yield of caffeic acid,total phenolic acid and total flavonoid as the evaluation indexes.Results The optimized parameters of the extraction by water are as follows:12 times of water and extract for 1.5 h in the first time,extraction for 1.0 h with 10 times of water in the second and the third time.The optimized parameters of the purification are as follows:the density is 1.05,alcohol concentration is 70%,standing time is 12 h.Conclusion The extraction and purification process of TH are stable and reasonable and feasible.
