摘要
目的探讨10-11-易位-甲基胞嘧啶双加氧酶(TETs)在结直肠癌中的表达及TET2对结直肠癌细胞增殖和侵袭的影响。方法利用GEPIA和UALCAN数据库分析TETs在结直肠癌组织中的表达差异;通过实时定量聚合酶链反应(qRT-PCR)、Western blotting和免疫组织化学方法检测TETs在结直肠癌组织和细胞中的表达;分别用CCK-8和侵袭实验检测TETs对结直肠细胞癌增殖和侵袭的影响。结果 TET2在结直肠癌组织和细胞系中低表达(P<0.05),TET1和TET3表达无差异;用质粒过表达TET2后,结直肠癌细胞的增殖和侵袭能力被显著抑制。结论 TET2在结直肠癌中显著低表达,且抑制结直肠癌细胞的增殖和侵袭,有望为结直肠癌的诊治提供新的靶点。
Objective To investigate the expression of ten-eleven translocation methylcytosine dioxygenases(TETs) in colorectal cancer and the effect of TET2 on the proliferation and invasion of colorectal cancer cells. Methods GEPIA and UALCAN databases were used to analyze the expression of TETs in colorectal cancer tissues. Real time quantitative polymerase chain reaction(qRT-PCR), Western blotting and immunohistochemistry(IHC) were used to detect the expression of TETs in colorectal cancer tissues and cells. CCK-8 and invasion assay were used to detect the effect of TETs on proliferation and invasion of colorectal cancer cells. Results TET2 was down-regulated in colorectal cancer tissues and cell lines(P<0.05). There was no significant difference in the expressions of TET1 and TET3. Overexpression of TET2 significantly inhibited the proliferation and invasion ability of colorectal cancer cells. Conclusion TET2 is significantly low expression in colorectal cancer and inhibits the proliferation and invasion of colorectal cancer cells, which is expected to provide a new target for the diagnosis and treatment of colorectal cancer.
