摘要
为了优化黄精总蛋白的提取工艺,试验采用碱液提取盐酸沉淀的方法提取黄精总蛋白,考马斯亮蓝法测定黄精蛋白含量。以黄精蛋白提取率为指标,将提取液的料液比、提取时间、提取次数、pH作为单因素来考察,通过单因素试验确定各因素水平。在单因素试验的基础上,利用响应面分析法优化碱提酸沉法提取黄精蛋白的最佳工艺。采用高效液相色谱法测定黄精中氨基酸种类。结果表明,最佳条件为料液比1︰5.6 (g/mL)、提取时间3.5 h、提取次数2次、pH 9.7。在这个最佳条件下进行3次验证性试验,蛋白质提取率的平均值为0.51%,与理论值相比较为接近,表明该数学模型可用于优化黄精蛋白提取工艺。同时采用高效液相色谱法分析黄精中所含的氨基酸种类,证明黄精中有多种对人体有益氨基酸。
To figure out the best extraction method of sealwort protein, proteins were extracted from sealwort by alkali extraction and acid precipitation, and coomassie blue staining was applied to calibrate the protein concentration within the extract. The best protein extraction method was figured out by response surface methodology and mono-factorial experiment on 4 factors which were liquid rate, extraction time, extraction times and pH in 3 levels for each. Based on the single factor experiment, the optimal process for extracting sealwort protein by alkali extraction and acid precipitation was optimized by response surface methodology. The amino acid species in sealwort were determined by high performance liquid chromatography. The results showed that the optimal conditions for polygonatum protein extraction was 1︰5.6(g/mL) of liquid rate, 3.5 h of extraction time, 2 times of extraction times and 9.7 of pH. Under this condition, the yield of polygonatum protein was 0.51%. Optimized regression equation derived some practical significance. Analysis of amino acid species contained in polygonatum by high performance liquid chromatography. It was proved that there were many kinds of beneficial amino acids in the essence of sealwort.
