摘要
目的研究采用代谢组学方法,探索补中益气汤改善脾虚型大鼠胃肠动力学障碍的潜在作用机制。方法采用LC-LTQ-OBITRAP高分辨质谱方法检测正常、模型及补中益气汤组血清代谢物变化,分析补中益气汤改善胃肠动力学障碍的血清生物标记物,解析补中益气汤作用的内源性代谢通路。结果研究发现补中益气汤对脾虚胃肠动力障碍大鼠体内的代谢轮廓有显著影响,与正常组相比,脾虚胃肠动力障碍大鼠有24种差异代谢物发生显著变化,而补中益气汤给药后可逆转其中14种差异代谢物,包括色氨酸、谷氨酰胺、磷脂胆碱等,通路分析研究表明,这些差异性代谢物主要与谷氨酰胺代谢、初级胆汁酸生物合成,不饱和脂肪酸生物合成、牛磺酸和亚牛磺酸代谢、丙氨酸和天冬、谷氨酸代谢有关。结论补中益气汤改善脾虚型大鼠胃肠动力学障碍可能与其影响代谢物轮廓,调控谷氨酰胺代谢、初级胆汁酸生物合成、不饱和脂肪酸生物合成、氨基酸代谢等有关。
Objective This study used metabolomics to explore the potential mechanism of Buzhong Yiqi Decoction improving gastrointestinal motility of the rats with spleen deficiency.Methods LC-LTQ-OBITRAP high-resolution mass spectrometry was used to detect the changes of serum metabolites in normal,model and Buzhong Yiqi Decoction groups.We found the endogenous metabolic pathways by analyzing the serum biomarkers of Buzhong Yiqi Decoction improving gastrointestinal dysfunction.Results The study found that Buzhong Yiqi Decoction had a significant effect on the metabolic profile of spleen-deficiency gastrointestinal motility disorder rats.Compared with the normal group,there were 24 significant metabolites in the spleen-deficiency gastrointestinal motility disorder.After the administration of Buzhong Yiqi Decoction,14 different metabolites,including tryptophan,glutamine,phospholipid choline,etc,can be reversed.Pathway analysis showed that these differential metabolites were mainly related to the biosynthesis of primary bile acid,metabolism of glutamine,biosynthesis of unsaturated fatty acid,metabolism of taurine and hypotaurine,metabolism of alanine,aspartate and glutamate.Conclusion Buzhong Yiqi Decoction can improve the gastrointestinal dysfunction in rats with spleen deficiency,which may be related to the influence of metabolite profile,regulation of glutamine metabolism,primary bile acid biosynthesis,unsaturated fatty acid biosynthesis and amino acid metabolism.
