摘要
目的基于网络药理学方法探讨八珍汤抗癌的作用机制。方法通过ETCM获得八珍汤药物化学成分,采用SwissTargetPrediction预测化学成分的潜在靶点并于OMIM、TTD、DiGSeE、Drug Bank、以及HPO 5个疾病靶点数据库进行比对,获得八珍汤抗癌的作用靶点。采用DAVID对八珍汤抗癌作用靶点进行KEGG通路分析,最终通过Cytoscape 3.2.1构建八珍汤抗癌"活性成分-作用靶点-通路"网络,并筛选出degree排名前10的靶点。结果从八珍汤共筛选出310个活性成分,361个与抗癌相关的作用靶点。经过KEGG通路富集分析,参与了癌症途径、HIF-1信号通路、PI3K-Akt信号通路、T细胞受体信号通路、VEGF信号通路等通路。结论八珍汤抗癌的作用机制体现了中药多成分-多靶点-多途径的特点,为阐释其抗癌作用机制与物质基础提供参考依据。
Objective To study the mechanism of Bazhen Decoction for anti-cancer based on network pharmacology.Methods The main active components of Bazhen Decoction were obtained from the ETCM database.The potential targets of the active ingredients were predicted by SwissTargetPrediction and compared with the five disease target data bases including OMIM,TTD,DiGSeE,Drug Bank and HPO to obtain the target of Bazhen Decoction for anti-cancer.The KEGG pathway was analyzed by DAVID for the anti-cancer target of Bazhen Decoction.Finally,Cytoscape 3.2.1 was used to construct the anti-cancer"active ingredient-target-way"network of Bazhen Decoction,and the top 10 targets of degree were selected.Results A total of 310 active ingredients and 361 anti-cancer related targets were screened from Bazhen Decoction.After KEGG pathway enrichment analysis,it is involved in Pathways in cancer,HIF-1 signaling pathway,PI3 K-Akt signaling pathway,T cell receptor signaling pathway,VEGF signaling pathway and other pathways.Conclusions The anti-cancer mechanism of Bazhen Decoction reflects the characteristics of multi-component-multi-target-multi-pathway of traditional Chinese medicine.The results of this study provide a reference for explaining its anti-cancer mechanism and material basis.
