摘要
目的探索EB病毒潜伏感染和活动性感染的患者体内炎症反应情况,阐明EB病毒感染状态与机体免疫炎症反应的关系。方法EB病毒阳性的p3HR-1细胞系用TPA/NaB诱导不同时间(0、6、12、24、36、48 h)使其进入裂解复制,另外收集69例健康志愿者血浆和外周血单个核细胞,通过实时荧光定量PCR以及酶联免疫吸附实验(ELISA),比较白细胞介素-1β(IL-1β)的分泌情况以及肿瘤坏死因子-α(TNF-α)的表达情况。结果细胞水平实验结果显示IL-1β的分泌以及TNF-α的表达随诱导时间的增加而增加。69例健康志愿者中,有34例呈EB病毒阳性,阳性率为49.28%;其中潜伏感染者21例(30.43%),活动性感染者13例(18.84%)。相比于未感染者[(10.34±1.16)pg/mL、(7.62±4.20)],潜伏感染者血浆中IL-1β的分泌(11.50±1.24)pg/mL以及TNF-α的表达量(4.35±2.51)无明显变化,但活动性感染者血浆中IL-1β的分泌(32.59±13.07)pg/mL以及TNF-α的表达量(412.40±261.10)明显增加,差异有统计学意义(P<0.05)。结论EB病毒潜伏感染者体内炎症反应并未被激活,只有在活动性感染患者体内病毒进行裂解复制炎症因子的表达和分泌才会被大量激活。
Objective To investigate the inflammatory response of EB virus in the latent infection and EB virus reactivation,and clarify the relationship between EB virus and innate immunity during each stage of EB virus infection.Methods P3 HR-1 cells were induced by TPA and butyrate at different times(0,6,12,24,36,48 h)to enter the virus lytic cycle.The plasma and peripheral blood mononuclear cell of 69 healthy volunteers at different infection stages were collected.The secretion of extracellular interleukin 1β(IL-1β)and the expression of tumor necrosis factorα(TNF-α)at different time or different infection periods were determined by Real-time quantitative PCR and enzyme linked immunosorbent assay.Results Cell experiments show that the secretion of extracellular IL-1βand the expression of TNF-αincreased with the increase of induction time.Among 69 healthy volunteers,34 cases of EB virus positive were detected and the positive rate was 49.28%.Of them,21 were people with latent EB virus infection and 13 with lytic EB virus infection.Compared with the uninfected people[(10.34±1.16)pg/mL,(7.62±4.20)],the secretion of IL-1β(11.50±1.24)pg/mL and the expression of TNF-α(4.35±2.51)in the plasma of people with latent EB virus infection did not change significantly by Real-time quantitative PCR and enzyme linked immunosorbent assay.But the levels of IL-1β(32.59±13.07)pg/mL and TNF-α(412.40±261.10)of people with lytic EB virus infection were significantly higher than those in the uninfected group(P<0.05).Conclusion The expression levels of inflammatory factors in serum of people with lytic EB virus infection were higher than those of people with latent EB virus infection.
