摘要
目的:比较竹节参中4种主要皂苷成分竹节参皂苷Ⅴ、伪人参皂苷RT1、竹节参皂苷Ⅳ、竹节参皂苷Ⅳa及其组合物的降脂活性。方法:采用油酸诱导的HepG2细胞脂肪堆积模型,以辛伐他汀和血脂康为阳性对照,以三酰甘油(TG)清除率为指标,考察竹节参皂苷Ⅴ、伪人参皂苷RT1、竹节参皂苷Ⅳ、竹节参皂苷Ⅳa及其不同比例组合物的体外降脂活性差异。构建蛋黄乳诱导的小鼠急性高脂血症模型,以辛伐他汀和血脂康为阳性对照,以血清TG、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)为指标,比较4种皂苷成分组合物与竹节参总皂苷的体内降脂作用。结果:体外实验结果显示,在相同剂量下,4种皂苷成分组合物的降脂活性优于各竹节参皂苷单体化合物和竹节参总皂苷,TG清除率显著升高(P<0.05,P<0.01);其中竹节参皂苷Ⅴ、伪人参皂苷RT1、竹节参皂苷Ⅳ、竹节参皂苷Ⅳa按2∶1∶1∶1比例组合物的降脂作用最为显著。体内实验结果显示,上述配比的竹节参皂苷组合物可显著降低急性高脂血症小鼠血清TC、TG、LDL-C水平(P<0.05,P<0.01),升高HDL-C水平(P<0.05,P<0.01),且其降脂作用优于竹节参总皂苷。结论:竹节参皂苷Ⅴ、伪人参皂苷RT1、竹节参皂苷Ⅳ、竹节参皂苷Ⅳa按2∶1∶1∶1组合可发挥协同降脂作用。
Objective: To compare the hypolipidemic effects of chikusetsusaponin Ⅴ, pseudoginsenoside RT1, chikusetsusaponin Ⅳ, chikusetsusaponin Ⅳa(four main saponins in Panacis Japonici Rhizoma) and their combinations. Methods: The lipid accumulation model in vitro was induced in HepG2 cells by oleic acid. Taken simvastatin and Xuezhikang as positive controls and triglyceride clearance rate as an indicator, the hypolipidemic activity of the above four components and their combinations with different proportions in vitro was observed. The acute hyperlipidemia model was established in mice by intraperitoneal injection of yolk milk. Simvastatin and Xuezhikang were taken as positive controls, and the serum levels of total cholesterol(TC), triglyceride(TG), low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C) were used as indicators, the hypolipidemic effects of chikusetsusaponin combinations and total saponins from Panacis Japonici Rhizoma in vivo were compared. Results: The results of experiments in vitro showed that, at the same dose, the hypolipidemic activity of the four chikusetsusaponins combination was better than that of single chikusetsusaponin component and total saponins from Panacis Japonici Rhizoma, and the triglyceride clearance rate was significantly increased(P<0.05,P<0.01). The combination of chikusetsusaponin Ⅴ,pseudoginsenoside RT1,chikusetsusaponin Ⅳ and Ⅳa(2∶1∶1∶1) showed the best hypolipidemic effect. The results of experiments in vivo showed that,the combination of chikusetsusaponins at above ratio could significantly reduce the serum levels of TC,TG,LDL-C(P<0.05,P<0. 01) and increase the level of HDL-C(P<0.05,P<0.01) in mice with acute hyperlipidemia,and its hypolipidemic effect was better than that of total saponins from Panacis Japonici Rhizoma. Conclusion: The combination of chikusetsusaponin Ⅴ,pseudoginsenoside RT1,chikusetsusaponin Ⅳ and Ⅳ a(2∶1∶1∶1) can play the synergistic role in hypolipidemic effect.
