摘要
为研究砷暴露对小鼠心肌组织损伤及细胞凋亡的影响,试验选用40只5周龄C57BL/6小鼠,随机分为4组(对照组饮用去离子水,低、中、高剂量砷暴露组分别饮用含0.15、1.50、15.00 mg/L三氧化二砷的去离子水),90 d后检测各组心脏脏器系数、血清心肌酶谱活力、心肌细胞凋亡率及凋亡通路相关基因的表达。结果表明,不同浓度砷暴露组的心脏脏器系数均有所下降,其中,中、高剂量组与对照组相比差异显著;与对照组相比,中、高剂量组血清肌酸激酶(CK)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)、α-羟基丁酸脱氢酶(HBDH)活力显著升高,低剂量组各项指标变化不明显。TUNEL法检测显示,对照组中未发现有凋亡心肌细胞,各剂量砷暴露组心内膜和心外膜下均可见不同数量的凋亡心肌细胞分布。与对照组相比,心肌组织Bax的表达在中、高剂量组显著上调;Bcl-2的表达在中、高剂量组显著下调;Caspase-3的表达在高剂量组显著上调。亚慢性砷暴露对小鼠心肌造成特异性损伤,且随着染毒剂量的增加损伤程度加重;诱导心肌细胞凋亡是砷暴露致心脏毒性的重要机制。
To study the effects of arsenic exposure on myocardial injury and cardiomyocyte apoptosis,forty C57BL/6 mice aged 5 weeks were randomly divided into four groups.The mice in the control group were given deionized water,and the mice in the low-dose,medium-dose,and high-dose arsenic exposed groups were given deionized water containing 0.15,1.50,15.00 mg/L arsenic trioxide,respectively.After 90 d exposure,heart organ coefficients and myocardial enzyme activities in the serum were measured,cardiomyocyte apoptosis rate and the expression of genes related to apoptosis were detected.The results showed that the heart organ coefficients of each arsenic exposed groups decreased.Compared with the control group,the changes were significant in the medium-dose group and high-dose group.Compared with the control group,serum CK,AST,LDH and HBDH activities in the medium-and high-dose groups significantly increased.But there was no significant difference between the low-dose group and the control group.There was no apoptotic cardiomyocytes in the control group.But in each dose of arsenic exposed group,there were many apoptotic cardiomyocytes under the endocardium and epicardium.Compared with the control group,the expression of Bax was significantly up-regulated,and the expression of Bcl-2 was significantly down-regulated in the medium-and high-dose arsenic exposed groups.The expression of Caspase-3 was significantly up-regulated in the high-dose group.In a conclusion,subchronic arsenic exposure induces myocardial injury in mice.The extent and degree of damage is augmented with the increase of arsenic exposure dose.Apoptosis is an important mechanism of cardiotoxicity caused by arsenic exposure.
