摘要
目的探讨壮药玉叶金花药材抗炎镇痛作用的谱效关系。方法建立小鼠耳肿胀、扭体模型考察玉叶金花的抗炎、镇痛活性;采用HPLC法建立玉叶金花的指纹图谱;采用灰度关联分析法分析不同批次玉叶金花指纹特征峰与其抗炎、镇痛作用之间的谱效相关性。结果HPLC指纹图谱显示玉叶金花有26个共有峰,与抗炎作用的关联度为0.7272~0.5779,对抗炎作用贡献较大的峰号为19>2>3>23>25>8,其中25号峰为异绿原酸A,8号峰为新绿原酸;与镇痛作用的关联度为0.7333~0.5708,对镇痛作用贡献较大的峰号为5>11>20>4>16>17,其中11号峰为山栀苷甲酯,20号峰为8-O-乙酰山栀苷甲酯。结论玉叶金花的抗炎镇痛作用是多种化学成分共同作用的结果,为探索玉叶金花药材抗炎镇痛的药效物质基础与完善其质量控制提供了参考。
Aim To explore the spectrum-activity relationship anti-inflammatory and analgesic effect of Mussaenda pubescens Ait.f..Methods The anti-inflammatory and analgesic effects of Mussaenda pubescens Ait.f.were studied by ear swelling and writhing model,respectively.Fingerprints of Mussaenda pubescens Ait.f.were established by HPLC.Then the spectrum-activity relationship of different batches of Mussaenda pubescens Ait.f.was evaluated by grey correlation analysis method.Results There were 26 peaks in these fingerprints,among which the correlation degree of anti-inflammatory and analgesic activity was 0.7272~0.5779 and 0.7333~0.5708,respectively.Peak number 19,2,3,23,25(Isochlorogenic acid A),8(neochlorogenic acid)contributed more to anti-inflammatory effect,and peak number 5,11(Shanzhiside methyl ester),20(8-O-Acetyl Shanzhiside methyl ester),4,16,17 contributed more to analgesic effects.Conclusions Many chemical substances are involved in the anti-inflammatory and analgesic effects of Mussaenda pubescens Ait.f.,which provides a reference for exploring the active substances in anti-inflammatory and analgesic effects and improving the quality control.
