COX-2和MMP-9在子痫前期胎盘组织中的表达及意义 被引量:3

摘要 目的探讨子痫前期胎盘组织中环氧合酶2(COX-2)和基质金属蛋白酶9(MMP-9)的表达水平及其相关性。方法选择2016年2月至2018年3月在本院进行剖宫产的51例正常妊娠孕妇、55例轻度子痫前期孕妇和48例重度子痫前期孕妇作为研究对象。采用RT-PCR检测受试孕妇胎盘组织中COX-2和MMP-9 mRNA表达水平,Western blotting检测胎盘组织中COX-2和MMP-9蛋白表达水平,比较各组孕妇胎盘组织中COX-2和MMP-9表达水平差异性,分析子痫前期胎盘组织中COX-2和MMP-9表达相关性。结果轻度和重度子痫前期组产妇胎盘组织中COX-2 mRNA和蛋白表达水平均显著高于正常组(P<0.05),MMP-9 mRNA和蛋白表达水平均显著低于正常组(P<0.05);重度子痫前期组产妇胎盘组织中COX-2 mRNA和蛋白表达水平均显著高于轻度子痫前期组产妇(P<0.05),MMP-9 mRNA和蛋白表达水平均显著低于轻度子痫前期组产妇(P<0.05);子痫前期产妇胎盘组织中COX-2和MMP-9表达呈显著负相关关系(r=-0.787,P<0.001)。结论子痫前期胎盘组织中COX-2表达升高,而MMP-9表达降低,与子痫前期发生及病情严重程度有关,两者呈显著负相关关系。 Objective To investigate the expression levels and correlation of circulating oxygenase 2(COX-2)and matrix metalloproteinase 9(MMP-9)in placenta tissues of preeclampsia.Methods From February,2017 to March,2018,51 pregnant women with normal pregnancy,55 cases of mild preeclampsia pregnant women and 48 cases of severe preeclampsia pregnant women were selected as study subjects.The expression levels of COX-2 and MMP-9 mRNA in placenta tissues of pregnant women were detected by RT-PCR.Western blotting was used to detect the expression levels of COX-2 and MMP-9 proteins in placenta tissues.The differences of COX-2 and MMP-9 expression levels in placenta tissues of pregnant women were then compared,and the correlation between COX-2 and MMP-9 expressions in placenta tissues of preeclampsia was analyzed.Results The expression levels of COX-2 mRNA and protein in placenta tissue of the mild and severe preeclampsia groups were significantly higher than those of the normal group(P<0.05),while MMP-9 mRNA and protein expression levels were significantly lower than those in the normal group(P<0.05).The expression levels of COX-2 mRNA and protein in placenta tissues of the severe preeclampsia group were significantly higher than those of the mild preeclampsia group(P<0.05),while MMP-9 mRNA and protein expression levels were significantly lower than those in the mild preeclampsia group(P<0.05).There was a significant negative correlation between COX-2 and MMP-9 expressions in placenta tissues of preeclampsia(r=-0.787,P<0.001).Conclusion The expression of COX-2 in placenta tissue of preeclampsia is increased,while the expression of MMP-9 is decreased.These two markers are correlated with the incidence of preeclampsia and the severity of the disease,and there is a significant negative correlation between the two.
