摘要
乙酰化丁香酸及其衍生物是重要的医药中间体,其合成与应用是目前医药领域的研究热点。本文在以二氯乙烷为溶剂,三乙胺存在下研究了丁香酸的乙酰化反应。通过NMR、GC/MS、LC/MS、IR和UV等对相应产物进行结构剖析,发现在羟基乙酰化的同时,也发生了羧基的酯化反应,生成主产物3,5-二甲氧基-4-乙酰基苯甲酸和副产物3,5-二甲氧基-4-乙酰基苯甲酸-(2-氯乙基)酯。推测二氯乙烷和三乙胺以季铵盐的形式,协同催化生成副产物。
Syringic acid and its derivatives were important pharmaceutical intermediates,which synthesis and application are the research hotspots in the medicine field.In the acetylation reaction of syringic acid in the presence of dichloroethane and triethylamine.The structure of the product was confirmed by spectroscopic methods such as NMR,GC/MS,LC/MS,IR and UV.Through structure analysis,it was found that,the hydroxyl groups acetylation and carboxyl esterification proceed simultaneously,generating the main product 4-acetyl-3,5-dimethoxybenzoic acid,the by-product 4-acetyl-3,5-dimethoxybenzoic acid-(2-chloroethyl) ester.It was speculated that dichloroethane and triethylamine in the form of quaternary ammonium salt,concerted catalysis to produce by-products.
