摘要
目的:探讨芪实益肾方对肾小球肾炎大鼠的保护作用及其可能的作用机制。方法:雄性SD大鼠50只,采用阿奇霉素制作肾小球肾炎大鼠模型,随机分为空白对照组、模型组、高剂量组、中剂量组、低剂量组。每组10只,灌胃给药8周,每2周眼底静脉取血,监测尿素氮(BUN)、肌酐(SCr);采用ELISA法检测大鼠白介素-2(IL-2)、白介素-6、(IL-6)、肿瘤坏死因子α(TNF-α)的表达。于末次给药后,取肾脏组织,进行HE染色,观察肾脏形态学变化;采用Western Blot方法,监测大鼠凋亡因子Bcl-2、Bax的表达。结果:芪实益肾方干预后,大鼠肾脏组织肾小球水肿明显减轻。与模型组比较,给药6周后BUN、SCr表达明显降低;给药8周后IL-2、IL-6和TNF-α表达明显降低。Bcl-2蛋白表达明显升高,Bax蛋白表达明显降低。结论:芪实益肾方能有效降低肾小球肾炎大鼠BUN、SCr、IL-2、IL-6、TNF-α的表达,其保护的机制可能与调节了Bcl-2、Bax的表达有关。
Objective:To investigate the protective effect of Qishi Yishen Recipe for rats with glomerulonephritis and its pos-sible mechanism.Methods:50male SD rats were randomly divided into blank controlled group,model group,high dose group,middle dose group and low dose group.10rats in each group were intragastrically administered for 8weeks.Blood was taken from the fundus vein every two weeks.Urea nitrogen(BUN)and creatinine(SCr)were monitored,and the expressions of interleukin-2(IL-2),interleukin-6(IL-6),tumor necrosis factor-alpha(TNF-alpha)were detected by ELISA.After the last adminis-tration,the kidney tissues were taken for HE staining to observe the morphological changes of the kidneys.Western blot was used to detect the expression of apoptotic factors Bcl-2and Bax in rats.Results:After the intervention of Qishi Yishen Recipe,the glomer-ular edema of kidney tissue in rats was significantly alleviated.Compared with the model group,the expression of BUN and SCr de-creased significantly after 6weeks of administration,while the expression of IL-2,IL-6and TNF-alpha decreased significantly after 8weeks of administration.The expression of Bcl-2protein was significantly increased and the expression of Bax protein was signifi-cantly decreased.Conclusion:Qishi Yishen Recipe can effectively reduce the expression of BUN,SCr,IL-2,IL-6and TNF-αin glomerulonephritis rats,and its protective mechanism may be related to the regulation of Bcl-2and Bax expression.
