电感耦合等离子体质谱法测定人体尿液中镉含量的研究 被引量:1

摘要 目的采用电感耦合等离子体质谱(ICP-MS)法测定清远市部分人群尿中镉(Cd)含量的研究。方法选取2018年1~12月清远市各大工厂体检健康的工人200例作为研究对象,留取其尿液。将所有尿液样本均分为两份,分别采用两种前处理方法,一组采用直接稀释法(1%硝酸20倍稀释)处理,另一组采用微波消解法进行前处理,处理完成进行ICP-MS分析,采用内标元素铟(115In)补偿基体效应,用标准模式测定Cd含量。结果ICP-MS测定标准曲线方程为Y=6566.8x+4928.1,相关系数为0.9975,线性良好,检测范围为0.1~20μg/L,符合要求;直接稀释法测得200例人群尿样中Cd含量为(2.87±1.78)μg/L,微波消解法测得(2.93±1.82)μg/L,两种方法测得结果比较,差异无统计学意义(P>0.05);男女间Cd元素含量比较,差异无统计学意义(P>0.05);Cd元素含量在<20岁,20~40岁,>40岁三组年龄段间呈递增趋势,差异有统计学意义(P<0.05)。结论电感耦合等离子体质谱法测得清远地区工厂健康人群尿中镉含量均处于正常范围内,且有随着年龄增长而递增的趋势。直接稀释法相比微波消解法,处理简单快速,适用于大批量尿样中元素含量的测定。 Objective To study the determination of some people’s urine cadmium(Cd)content in urine by inductively coupled plasma mass spectrometry(ICP-MS)in Qingyuan city.Methods From January 2018 to December 2018,200 healthy people in major factories of Qingyuan were selected as the research subjects to take urine.All urine samples were divided into two parts and two pretreatment methods were adopted.One group was treated with direct dilution method(20%dilution of 1%nitric acid),the other group was treated with microwave digestion method,after the treatment,ICP-MS analysis was completed,and the internal standard indium(115In)was used to compensate matrix effect.Results The standard curve equation of ICP-MS was Y=6566.8x+4928.1,the correlation coefficient was 0.9975,and the detection range was 0.1-20ug/L,which met the requirements.The content of Cd in 200 urine samples measured by direct dilution method was(2.87±1.78)ug/L,and that measured by microwave digestion method was(2.93±1.82)ug/L.There was no statistical difference between the two methods(P>0.05).There was no significant difference in the content of Cd between men and women(P>0.05).The content of Cd elements was increased in the three groups of less than 20 years old,20-40 years old,and over 40 years old,and the difference between the groups was statistically significant(P<0.05).Conclusion The cadmium content in urine of healthy people in Qingyuan area is measured by inductively coupled plasma mass spectrometry.Compared with microwave digestion method,direct dilution method is simple and fast,which is suitable for the determination of element content in a large number of urine samples.
