摘要
目的对1例输入性疟疾病例进行鉴定。方法采集病例外周血制作滤纸血样和血涂片。血涂片染色后镜检,同时进行疟疾快速诊断试剂盒(RDT)检测,滤纸血样进行PCR,PCR扩增片段测序后在GenBank中进行Blast分析。结果RDT检测提示非恶性疟的疟原虫阳性,血涂片镜检可见卵形疟原虫。PCR扩增出大小为880 bp的卵形疟特异条带,Blast分析显示与卵形疟原虫wallikeri亚种的cox3部分基因序列同源性为99.4%。结论依据RDT、镜检、PCR和序列分析等实验室检测结果,结合其流行病学资料,该例输入性疟疾病例确诊为贵州省首次由境外输入的卵形疟病例,且感染的疟原虫虫种是卵形疟原虫wallikeri亚种。
Objective To identify a case of imported malaria in Guizhou Province.Methods Blood samples and smears were collected from peripheral blood of patients.The blood smear was made and examined under microscope,and the malaria rapid diagnostic test(RDT)detection was performed at the same time.The filter blood sample was performed by PCR,and PCR amplified fragment sequencing was performed by Blast analysis in GenBank.Results The RDT detection indicated that parasites of the non-falciparum malaria were positive,and Plasmodium ovale was visible by blood smear microscopy.A 880 bp specific band of Plasmodium ovale was amplified by PCR.Blast analysis showed that part of the cox3 gene sequence was 99.4%homologous to the wallikeri subspecies of Plasmodium ovale.Conclusion According to laboratory test results such as RDT,microscopy,PCR,sequence analysis,combined with the epidemiological data,this imported malaria case was confirmed as the first imported case of Plasmodium ovale malaria in Guizhou Province,and the infected Plasmodium ovale parasite species were the wallikeri subspecies of Plasmodium ovale.
