栓皮栎幼苗叶片内源激素含量对切根胁迫的短期动态响应 被引量:4

Short-term dynamic responses of endogenous hormones in leaves to root pruning of Quercus variabilis seedlings

摘要 为了研究切根胁迫对栓皮栎幼苗叶片内源激素含量动态变化及生长的影响,探索内源激素在短期内调节林木根—冠互作关系中发挥的生理作用。本研究以1 a生栓皮栎幼苗为研究对象,设置1/2切根、1/3切根处理,不切根作为对照(CK),采用酶联免疫法(ELISA)测定切根后第2、15、30、60、90天的栓皮栎幼苗叶片中IAA、ABA、GA3、GA4和MeJA等内源激素含量的短期动态变化规律及幼苗生长状况。结果表明(1)随着切根时间的延长,切根比例越大,与CK组相比ABA上升幅度越大,随着根系的自我修复,切根组ABA含量与CK组趋于相同水平;(2)切根后第2天,1/2切根组含量迅速升高,显著高于CK组(P<0.05),之后切根组逐渐降低,直到第90天时切根组显著高于CK组(P<0.05);(3)对于GA3和GA4,2~60 d内,切根对栓皮栎幼苗叶片中GA3含量变化无明显影响,在第90天时,1/2切根组显著高于1/3切根组和CK组(P<0.05);在切根2~60 d内,切根组叶片中GA4含量高于CK组,整个过程各组含量呈现先升高后降低趋势;(4)切根后栓皮栎幼苗叶片中MeJA含量短期内逐渐升高,切根组含量均低于同期CK组,且1/2切根组高于1/3切根组。切根作为对根系的一种伤害,在短期内对各激素有一定影响,切根比例越大,激素含量上升幅度越大。 In order to study the effects of root-pruning stress on the growth and the dynamic changes of endogenous hormone levels in the leaves of Quercus variabilis seedlings,and to explore its physiological mechanism in the short term regulation of root-crown interaction,1 a Quercus variabilis seedlings were used as research objects by setting 3 treatment groups,1/2 root-pruning group,1/3 root-pruning group,and non-pruning of root as control group(CK).ELISA method was used to determine the law of short-term change of endogenous hormones(such as IAA,ABA,GA3,GA4,and MeJA)in leaves of Q.variabilis seedlings on the 2nd,15th,30th,60th and 90th day after root pruning,and the growth of the seedlings was analyzed as well.The results showed that:(1)With the extension of root pruning time,compared with the CK group,the larger the proportion of root pruning was,the greater the increase of ABA content.With the root self-repairing,the ABA content in the root-pruning group tended to be at the same level as that in the CK group;(2)From the 2nd day after root pruning,the content of IAA in the leaves of 1/2 root-pruning group increased rapidly,which was significantly higher than that in the CK group(P<0.05),then the content of IAA in the leaves of the root-pruning group gradually decreased until the 90th day,when it began to increase again to a significantly higher level than that of the CK group(P<0.05);(3)Root pruning had no significant effect on the change of GA3 content in the leaves of Quercus variabilis seedlings.On the 90th day,GA3 content in the leaves of seedlings in the 1/2 root-pruning group was significantly higher than that in the 1/3 root-pruning group and the CK group(P<0.05);within 2 to 60 days after root pruning,the GA4 content in the leaves of seedlings in the root-pruning group was higher than that in the CK group,and in the whole process,the content of each group showed the tendency of increasing first and then decreasing;(4)After root-pruning,theMeJA content in the leaves of seedlings gradually increased in a short period and was lower in both of the root-pruning groups than that in the CK group,and it was higher in the 1/2 root-pruning group than in the 1/3 root-pruning group.Root-pruning,as an injury to the root system,has a certain effect on each hormone in a short period of time.The larger the proportion of root-pruning is,the greater the increase of hormone content is.
