摘要
目的通过观察筋骨痛消丸对膝骨关节炎大鼠模型血清中MLT、MMP-3和MMP-13表达的影响,探究筋骨痛消丸治疗膝骨关节炎的机制,为临床防治膝骨关节炎提供实验依据。方法SPF级SD大鼠40只,体质量200~220 g,随机分为4组,空白对照组、模型对照组、阳性对照组、实验组,每组10只。除正常对照组外,其余各组大鼠均采用关节腔内注射木瓜蛋白酶溶液制备膝骨关节炎模型。造模后,按照人体用药量换算成大鼠等效剂量作为大鼠给药量,阳性对照组大鼠每天予以双醋瑞因药液给药,实验组每天予以筋骨痛消丸药液给药,空白对照组和模型对照组每天予以生理盐水给药,各组连续给药两周。于给药后采集血液样本,运用ELISA检测法检测各组大鼠血清中MLT及MMP-3、MMP-13的表达水平。结果给药后,与模型对照组相比,阳性对照组和实验组大鼠的关节肿胀度明显降低(P<0.05),大鼠血清中MMP-3、MMP-13的表达水平明显降低(P<0.05),且实验组降低显著(P<0.05);与其余各组相比,实验组大鼠血清中MLT的表达水平显著升高(P<0.05)。结论筋骨痛消丸可以上调膝骨关节炎大鼠模型血清中MLT的表达水平,降低MMP-3、MMP-13的表达水平,从而增强机体抗氧化、抗炎及抗凋亡等作用,对膝骨关节炎的治疗具有积极作用。
Objective To investigate the effect of Jingu Tongxiao Pill on the expression of MLT,MMP-3 and MMP-13 in serum of rats with knee osteoarthritis.To study the mechanism of Jingu Tongxiao pills in the treatment of knee osteoarthritis,To provide experimental and theoretical basis for clinical application of Jingu Tongxiao pills in the treatment of knee osteoarthritis.Methods Forty SPF grade SD rats weighing 200g-220g were randomly divided into four groups:blank control group,model control group,positive control group and experimental group.Except for the normal control group,all the rats in the other groups were injected with papain solution to make the model of knee osteoarthritis.After the establishment of the model,according to the amount of drugs used in the human body,the rats were converted into the equivalent dose of rats as the dosage of rats.The rats in the positive control group were given double vinegar Riin solution every day,and the experimental group was given Jingu Tongxiao pill every day.The blank control group and the model control group were given normal saline every day,and each group was given the drug for two weeks.Blood samples were collected after administration,and the expression levels of MLT,MMP-3 and MMP-13 in serum of rats in each group were detected by ELISA method.Results After administration,compared with the model control group,the joint swelling degree of the positive control group and the experimental group decreased signifi cantly(P<0.05),and the expression levels of MMP-3 and MMP-13 in the serum of the rats decreased signifi cantly(P<0.05).And the experimental group decreased signifi cantly(P<0.05).Compared with the other groups,the expression level of MLT in the serum of the experimental group was signifi cantly higher than that of the other groups(P<0.05).Conclusion Jingu Tongxiao Pill can up-regulate the expression of MLT and decrease the expression of MMP-3 and MMP-13 in the serum of knee osteoarthritis rats,so as to enhance the effects of antioxidation,anti-infl ammation and anti-apoptosis.It has a positive effect on the treatment of knee osteoarthritis.
