青钱柳对2型糖尿病大鼠早期肾损伤标志物以及氧化应激的影响 被引量:12

Effects of Cyclocarya paliurus on early kidney injury markers and oxidative stress in T2DM rats

摘要 目的探究青钱柳对2型糖尿病大鼠的中性粒明胶酶脂质运载蛋白(neutral gelatinase lipoprotein,NGAL)、肾损伤因子1(kidney damage factor 1,KIM-1)、铜蓝蛋白(ceruloplasmin,CP)以及氧化应激的影响。方法雄性SD大鼠100只,适应性喂养1周,将大鼠随机分为空白组10只和造模组90只。其中空白组给予普通饲料,造模组给予高脂饲料喂养8周。第10周,以30 mg/kg的剂量一次性腹腔注射2%链脲佐菌素(streptozotocin,STZ)(现配现用,避光取适量STZ,在0.1 mol/L柠檬酸-柠檬酸钠缓冲液冰浴中配置,pH=4.5)。72小时后选择空腹血糖>11.1 mmol/L的成模大鼠随机分为模型组,二甲双胍组,青钱柳水提物(Cyclocarya paliurus water extract,CPE)低、中、高剂量组,每组10只。空白组和模型组给予0.9%氯化钠溶液,二甲双胍组给予二甲双胍混悬液(0.3 g/kg),CPE低、中、高剂量组分别给予青钱柳叶水提取物[CPE(0.25、0.5、1 g/kg)]。连续给药4周,期间检测空腹血糖(fasting blood glucose,FBG)。给药4周后,检测肌酐(creatinine,CRE)、尿素氮(urea nitrogen,UREA)、NGAL、KIM-1、CP、丙二醛(malondialdehyde,MDA)、活性氧(reactive oxygen species,ROS)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)、过氧化氢酶(catalase,CAT)、超氧化物歧化酶(superoxide dismutase,SOD)的含量。结果与模型组比:(1)CPE低、中、高剂量组FBG显著降低(P<0.05);(2)CPE低、中、高剂量组CRE、UREA、NGAL、KIM-1、CP显著降低(P<0.05);(3)CPE低、中、高剂量组MDA、ROS显著降低,GSG-PX、CAT、SOD显著升高(P<0.05)。结论青钱柳可有效缓解氧化应激损伤,从而防治2型糖尿病早期肾损伤。 Objective To explore the effect of Cyclocarya paliurus on early kidney injury markers and oxidative stress in T2DM rats,the markers includes neutral gelatinase lipoprotein(NGAL),kidney damage factor 1,(KIM-1)and ceruloplasmin(CP).Methods A total of 100 male Sprague-Dawley rats were randomly fed for one week.The rats were randomly divided into a normal group of 10 and a model group of 90.The normal group was given normal feed,and the model was given high fat diet for 8 weeks.In the 10th week,a 2% streptozotocin(STZ)was intraperitoneally injected at a dose of 30 mg/kg(currently available,appropriate amount of STZ in the dark,0.1 mol/L citric acid-sodium citrate in the buffer ice bath,pH=4.5),and after 72 h,the model rats with fasting blood glucose>11.1 mmol/L were randomly divided into model group,metformin group,CPE low,medium and high dose group,10 per group;The normal group and model group were given 0.9% sodium chloride solution,the metformin group(0.3 g/kg),and the CPE low,medium and high dose group were given respectively CPE(0.25,0.5,1 g/kg).During 4 weeks’continuous gavage,blood in tail vein was collected to measure fasting blood glucose(FBG).After 4 weeks’treatment,creatinine(CRE),urea nitrogen(UREA),neutral gelatinase lipoprotein(NGAL),kidney damage factor 1(KIM-1),ceruloplasmin(CP),malondialdehyde(MDA),reactive oxygen species(ROS),glutathione peroxidase(GSH-PX),catalase(CAT),superoxide dismutase(SOD)were measured.Results Compared with the model group:(1)FBG was significantly decreased in the low,medium and high dose group of CPE(P<0.01,P<0.05);(2)CRE,UREA,NGAL,KIM-1,CP was significantly decreased in the low,medium and high dose groups of CPE(P<0.01,P<0.05);(3)MDA and ROS were significantly decreased and GSG-PX,CAT and SOD were significantly increased in the low,medium and high dose group of CPE(P<0.01,P<0.05).Conclusion Cyclocarya paliurus can effectively regulate oxidative stress reaction and prevent early kidney injury.
