青海省海北州不同地区蜱及蜱传病原的分子生物学鉴定 被引量:5

Identification of Ticks and Tick-borne Pathogens in Different Areas of Haibei of Qinghai Province

摘要 为了阐明青海海北地区患病藏羊携带蜱虫种类及蜱传病原的情况,采用PCR方法对蜱虫种类进行鉴定,并利用16S rRNA基因进行遗传进化分析,同时也利用PCR方法检测了携带的病原,对鉴定的病原进行分析。结果显示:青海海北地区主要流行西藏革蜱、森林革蜱、青海血蜱和草原革蜱,鉴定的蜱虫基因与GenBank中的参考基因的同源性均在97.00%~100.00%之间;遗传进化分析显示鉴定的蜱虫均与相应的蜱种聚类,且均与西北地区的分离株聚在同一分支上,呈现区域聚集性;蜱传病原种类丰富,包括尤氏泰勒虫、吕氏泰勒虫、绵羊泰勒虫、巴尔通体、绵羊无浆体、立克次体、贝纳特氏立克次体,流行的主要病原是绵羊无浆体和巴尔通体。通过本次的分子生物学鉴定研究,有助于研究我省蜱的生物学多样性和进化关系,丰富蜱种分子标记相关基因序列信息;同时对蜱传病原进行了分子鉴定,有助于查清蜱携带的病原谱,可为我省蜱传病的防控提供科学依据。 In order to investigate the ticks species and tick-borne pathogens in the sick Tibetan sheep in Haibei area of Qinghai,some ticks samples were collected during the diagnosis of animal diseases.The PCR method was used to identify ticks species,and 16 S rRNA gene was used for phylogenetic analysis.The tick-borne pathogens were also detected and analyzed by PCR method.The results showed that Dermacentor everestianus,Haemaphysalis qinghaiensis,Dermacentor silvarum and Dermacentor nuttalli were prevalent in Haibei of Qinghai.The identified ticks genes were homologous with reference genes in GenBank between 97.00%and 100.00%.The phylogenetic analysis showed that the identified ticks were clustered with the corresponding ticks species isolated from the northwestern region of China,presenting regional clustering.The species of tick-borne pathogens were abundant,included Theileria uilenbergi,Theileria luwenshuni,Theileria ovis,Bartonella schoenbuchensis,Anaplasma ovis,Rickettsia raoultii,Coxiella burnetii,Anaplasma ovis and Bartonella schoenbuchensis were the predominant pathogens.This molecular identification study is helpful to study the biological diversity and evolutionary relationship of ticks in Qinghai province,and to enrich the sequences information of ticks marker related genes.The results of molecular identification of tick-borne pathogens are helpful to investigate the spectrum of pathogens carried by ticks.It can provide scientific basis for the prevention and control of tick-borne diseases in Qinghai Province.
