摘要
目的观察木香乙醇提取物对模拟高原缺氧小鼠肝脏线粒体相关生化指标的影响,从线粒体功能角度探讨其对肝脏的保护作用机制。方法Balb/c小鼠随机分为正常对照组、缺氧模型组、乙酰唑胺组、木香70%乙醇提取物低、中、高剂量组,每组10只。乙酰唑胺组给予腹腔注射300 mg/kg乙酰唑胺,低、中、高剂量组分别给予木香70%乙醇提取物125、250、375 mg/kg。给药后缺氧72 h麻醉处死,提取肝脏线粒体,测定线粒体膜电位、线粒体复合体Ⅰ、Ⅱ和苹果酸脱氢酶、顺乌头酸酶、α-酮戊二酸脱氢酶活性。结果木香70%乙醇提取物低、中、高剂量组线粒体膜电位、线粒体顺乌头酸酶、α-酮戊二酸脱氢酶活性和呼吸链复合体Ⅱ活性高于缺氧模型组,高剂量组线粒体苹果酸脱氢酶活性低于缺氧模型组,线粒体呼吸链复合体Ⅰ活性高于缺氧模型组(P<0.05,P<0.01)。结论木香70%乙醇提取物具有保护缺氧小鼠肝脏线粒体功能的活性。
Objective To observe the effect of ethanol extract of Saussurea costus on the mitochondria-related biochemical parameters of liver of mice exposed to simulated high-altitude hypoxia,and to explore its protective mechanism on liver from the perspective of mitochondrial function.Methods Balb/c mice were randomly divided into normal control group,hypoxia model group,Acetazolamide group,low,medium,and high-dose 70%ethanol extract groups,with 10 mice in each group.Acetazolamide group was given intraperitoneal injection of 300 mg/kg Acetazolamide,and low,medium and high-dose groups were given 70%ethanol extract of Saussurea costus(125,250 and 375 mg/kg respectively).At 72 h after exposure to hypoxia,they were anesthetized and sacrificed.Liver mitochondria was collected,and mitochondrial membrane potential,mitochondrial complex Ⅰ,Ⅱ and malate dehydrogenase,aconitase,and α-ketoglutarate dehydrogenase activity were measured.Results The mitochondrial membrane potential,and activities of mitochondrial aconitase,α-ketoglutarate dehydrogenase and respiratory chain complexⅡin the low-,medium-and high-dose groups were higher than those in the hypoxia model group.The activities of mitochondrial malate dehydrogenase was lower,and mitochondrial respiratory chain complexⅠwas higher in the high-dose group than in the hypoxia model group(P<0.05,P<0.01).Conclusion 70%ethanol extract of Saussurea costus can protect liver mitochondrial function in hypoxic mice.
