干涉显微细胞相位快速恢复及图像预处理分析 被引量:1

摘要 和传统成像技术相比较,相位显微成像应用期间省略了染色环节,并且能实现无损伤性操作,成像结果可以数量化,近年来在生物细胞学科领域中有较广泛应用。很多学者陆续提出较先进的定量相位成像技术和光波干涉原理存在一定相关性。文章首先阐述了相移干涉显微成像基本原理及典型的相位恢复方法;其次提出新型微分相位提取法;最后对显微镜下细胞图像进行预处理,并以此方法作出模拟及实验验证,目的是阐述相位梯度用于辨识细胞类别及内部结构的可行性。 Compared with the traditional imaging technology, the staining link is omitted during the application of phase microscopic imaging, and the damage-free operation can be realized, and the imaging results can be quantified, which has been widely used in the field of biological cell science in recent years. Many scholars have put forward more advanced quantitative phase imaging technology, many of which have some correlation with the principle of light wave interference. Firstly, the basic principle and typical phase recovery method of phase-shifting interference microscopy are introduced in this paper. Secondly, a new differential phase extraction method is proposed. Finally, it preprocesses the cell image under the microscope, and uses this method to make simulation and experimental verification. The feasibility of using phase gradients to identify cell types and internal structures is described.
