摘要
【目的】蓝桉、巨桉和大叶桉是成都地区广泛引种栽培的绿化植物,本文挖掘了桉树植物资源的利用潜力。【方法】用水蒸气蒸馏法提取叶挥发油后,用气相色谱-质谱联用法(GC-MS)分析了其成分和相对含量,同时采用四甲基偶氮唑蓝比色法(MTT)评价了3种桉树叶挥发油的体外抗肿瘤活性。【结果】蓝桉、巨桉和大叶桉叶的挥发油得率分别为3.67%、1.21%和0.43%;分别从它们的叶挥发油中鉴定出34、34和31种化合物;α-蒎烯、莰烯、右旋柠檬烯、桉油精、葑醇、龙脑烯醛、龙脑、香芹酮、别香橙烯、石竹烯、邻伞花烃等共17种化合物是3种挥发油的共有成分。含量最高的成分均为α-蒎烯和桉油精,在蓝桉、巨桉、大叶桉叶挥发油中,α-蒎烯的相对含量分别为17.18%、17.89%、41.91%,桉油精的相对含量分别为59.55%、50.49%、7.68%;3种桉树叶挥发油对人肝癌SMMC-7721细胞和人正常肝L02细胞的增殖均具有一定的抑制作用,但对SMMC-7721细胞的抑制效应大于L02细胞;大叶桉、巨桉、蓝桉叶挥发油对SMMC-7721细胞的IC50为4.66、7.36、13.53μg·mL^-1,以大叶桉挥发油抗肿瘤活性最佳。【结论】同一地区桉属不同种类的挥发油组成成分和含量具有一定的差异,桉树挥发油具有开发为抗肿瘤药物的潜力。
【Objective】Eucalyptus globulus,E.grandis and E.robusta are green plants widely cultivated in Chengdu,and this paper aimed to exploit the utilization potential of Eucalyptus resources.【Method】The essential oils from three Eucalyptus species leaves were extracted by using the modified steam-distillation extraction method,and the chemical components were determined by gas chromatography-mass spectrometry(GC-MS).The effects of the essential oils on proliferation of human hepatoma cells(SMMC-7721)in vitro was studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT)assay.【Result】The oil yields of E.globulus,E.grandis and E.robusta were 3.67%,1.21%and 0.43%,respectively.The essential oil of E.globulus,E.grandis and E.robusta contains 34,34 and 31 components.A total of 17 components,such asα-pinene,camphene,limonene,eucalyptol,fenchol,campholenal,borneol,carvone,alloaromadendrene,caryophyllene and o-cymene,were common components of three essential oils.The two main components wereα-pinene and eucalyptol in the essential oil of E.globulus,E.grandis and E.robusta,the relative contents ofα-pinene were 17.18%,17.89%and 41.91%,respectively;The relative contents of eucalyptol were 59.55%,50.49%and 7.68%,respectively.Three Eucalyptus essential oil significantly inhibited proliferation of human hepatoma SMMC-7721 cells and human normal liver L02 cells.SMMC-7721 cells were more inhibited than L02 by the essential oil of E.robusta,E.grandis and E.globulus,with half maximal inhibitory concentration(IC50)values of 4.66,7.36 and 13.53μg·mL^-1 after 24 hours treatments,respectively.The essential oil from E.robusta had a best inhibiting effect on SMMC-7721 cells.【Conclusion】These results indicated that the chemical components and contents of essential oils from different species of Eucalyptus in the same area were different,and the essential oils of Eucalyptus had the potential to be used as antitumor drugs.
