摘要
甲烷氧化菌素(methanobactin,mb)是甲烷氧化菌分泌的一种可结合铜的小分子荧光肽,具有氧化还原活性.采用摇瓶法培养甲烷氧化菌,通过不同培养基装液量来寻找菌体的生长规律,确定最适生长条件,并采用大孔树脂法分离纯化甲烷氧化菌素,通过大孔树脂对甲烷氧化菌素的静态吸附及动态吸附方式对比,找到最佳的吸附效果.实验结果表明,摇瓶法培养的最佳培养基装液量1.5 L,通过大孔树脂静态吸附是最佳的吸附甲烷氧化菌素方式,为今后提高甲烷氧化菌素的分离能力奠定了科学基础.
Methanobactin(mb)was a small molecule fluorescent peptide that bond to copper from methanotrophs,and had redox activity.After methanotrophs were cultured by shake-flask fermentation,andthe optimum growth conditions were determinated by different volume of medium.According to the difference between static adsorption and dynamic adsorption of macroporous resin,the better adsorption effect was found.The results showed that the optimum volume of culture medium was 1.5 L,and the static adsorption of macroporous resin was the better way to obtainmethanobactin than dynamic adsorption.The results had proved useful information ofimproving methanobactin productivity.
