摘要
目的:探讨miRNA-30a-5p在肾癌细胞中的作用及其机制。方法:将miRNA-30a-5p模拟物及抑制物转染至肾癌786-0细胞。检测miRNA-30a-5p在各组细胞中的表达、细胞增殖能力、细胞凋亡情况及凋亡抑制蛋白(XIAP)表达。结果:miRNA-30a-5p mRNA的表达:模拟物组明显上升,未转染组、抑制物组明显下降;细胞增殖能力:各组均有随时间延长而升高的趋势;模拟物组凋亡率高于抑制物组及未转染组,XIAP蛋白的相对表达量低于抑制物组及未转染组。结论:miRNA-30a-5p过表达并通过靶向XIAP抑制肾癌786-0细胞的增殖,促进细胞凋亡。
Objective:To investigate the role of miRNA-30a-5p in renal cancer cells and its mechanism.Methods:miRNA-30a-5p mimics and inhibitors were transfected into renal cancer 786-0 cells.The expression of miRNA-30a-5p in each group of cells,cell proliferation ability,apoptosis and XIAP protein expression were detected.Results:The expression of miRNA-30a-5p mRNA was significantly increased in the simulant group,and decreased significantly in the non-transfected group and the inhibitor group.The cell proliferation ability:each group had a tendency to increase with time.The apoptosis rate in the mimic group was higher than that in the inhibitor group and non-transfected group,the relative expression of XIAP protein was lower than that of the inhibitor and non-transfected groups.Conclusion:miRNA-30a-5p is over-expressed and inhibits the proliferation of renal cancer 786-0 cells by targeting XIAP,and promotes apoptosis.
