摘要 [目的]本文旨在研究茄子灰霉病原菌多样性,为灰霉病防治提供依据。[方法]从运城保护地茄子灰霉病株上分离纯化了15个灰葡萄孢菌株,并对其进行了生物学表型和遗传学多样性分析。通过观察菌株的生长速率、菌核的大小、形状及分布明确其表型多样性,结合RAPD分子标记利用18条随机引物进行遗传多样性分析。[结果]借助ITS和PCF/R引物PCR扩增鉴定15个菌株均为灰葡萄孢(Botrytis cinerea)。各菌株在生长速率、菌核形成及分布方面存在明显差异。菌株QZBC4生长速率最慢,而菌株QZBC1、8和13生长速率较快达到1.4 cm·d-1。15个菌株的菌落类型有4种,菌丝型、孢子型、细小菌核型和大菌核型。筛选出的4条RAPD引物共扩增出条DNA条带30条,其中多态性条带为22条,多态性比率变化范围在63.6%~87.5%,平均值为73.3%。UPGMA聚类分析标明,在阈值0.76下,15个菌株可以分为4个类群。[结论]RAPD分析结果与表型分析结果一致,说明茄子灰葡萄孢菌株间存在明显的遗传多样性。 [Objective]The present study was conducted to investigate the diversity of gray mold pathogenson eggplants and to provide scientific support for gray mold control in the field.[Methods]In this study,15 strains were isolated from eggplants in Yuncheng city.The biological phenotype and genetic diversity of collected strains were analyzed.Conidial morphological features of each strain including the growth rate,conidial shape,and distribution of sclerotium were evaluated.Eighteen random selected primers were used as RAPD molecular markers to analyze genetic diversity of the pathogens.[Results]The ITS and BCF/R primers PCR amplification results indicated that all 15 isolates strains were belonged to B.cinerea.The obviously different biological phenotypes were observed within all strains,such as different growth rates,sclerotium formation and distribution.The slowest growth rate of pathogens was detected with strain QZBC4 at 0.5 cm·d-1,while the strains QZBC 1,8,and 13 produced greater growth rate at 1.4 cm·d-1.The colonies of 15 strains were divided into four types,including mycelia,spore,microsclerotia,and large sclerotia.Four RAPD selected primers produced 30 DNA bands in total,of which 22 DNA bands were polymorphic bands with the percentage of polymorphic DNA bands ranging from 63.6%to 87.5%,and the average value was 77.3%.UPGMA cluster analysis demonstrated that 15 isolates were divided into four groups at a threshold of 0.76.[Conclusion]RAPD results were consistent with that of phenotypic analysis,indicating that B.cinerea strains on eggplants possess obvious genetic diversity.
