摘要
采用土壤微生物稀释平板法分离技术,从烟草根际土壤分离到一株活体纯培养物,将其编号为ZMD1。ZMD1菌株与2种病原的拮抗试验均显示出一定的抗菌活性;通过聚酮合成酶(PKS)基因的兼并引物对ZMD1进行了克隆验证,测序结果与其他物种的PKS基因序列具有较高的相似性,故将ZMD1初步定性为产聚酮合成酶活性菌株。综合ZMD1菌株的一般形态特征、生理生化特征和菌株的16S rDNA保守序列特征分析,初步将ZMD1确定为链霉菌属(Streptomyces)菌株,暂命名为Streptomyces sp.ZMD1。ZMD1可作为烟草病害生防菌剂、土壤保育菌剂和专用菌肥等开发的良好备用材料。
A pure bacterial culture was isolated from tobacco rhizosphere soil by plate culture technique,and its number was ZMD1.ZMD1 strain showed some antagonistic activity against two pathogens.ZMD1 was identified as a polyketide synthase producing strain by cloning with the merger primers of the polyketone synthase(PKS)gene,and the results of sequencing were similar to those of PKS gene sequences of other species.Based on the general morphological characteristics,physiological and biochemical characteristics and 16S rDNA conservative sequence analysis of ZMD1 strain,ZMD1 strain was initially identified as Streptomyces strain,tentatively named Streptomyces sp.ZMD1.ZMD1 strain can be used as a good reserve material for the development of biocontrol agents for tobacco diseases,soil conservation agents and special fertilizer.
