摘要
为了获得对致病菌Canidia albicans YTS-03具有强抗性的嗜杀酵母工程菌,以开发抗真菌制剂。对所筛选的4株优良菌株PA1、SY3-1A、DD21-2、WM1进行了耐高温性、耐酸碱性、耐盐性、抗生素抗性及抗菌能力检测。结果表明:PA1、SY3-1A、DD21-2、WM1对温度和环境pH的适应范围较宽,可以在12~37℃及pH3~9的条件下生长繁殖,并且其耐盐性存在较大的差异,分别为10%、8%、7%和4%;温度、pH和盐度均对实验菌株生长有显著影响(P<0.05)。4株菌对抗菌素较为敏感,对伊曲康唑和低浓度(5μg/mL)的制霉菌素或低浓度(5μg/mL)的氟康唑有抗性,并且4株嗜杀酵母菌株具有不同的抗药性特征。另外,4株嗜杀酵母菌株PA1、SY3-1A、DD21-2、WM1均能杀死白色假丝酵母、二尖梅奇酵母和热带假丝酵母,而且表现出高嗜杀活力,其菌液嗜杀因子对C.albicans YTS-03菌株的抑菌圈直径分别达到23.0、14.1、22.3和21.5 mm。
In order to obtain an engineered killer yeast which was highly resistant to the fungal pathogen Canidia albicans YTS-03,and to develop antifungal preparations,four kinds of excellent strains for resistance of high temperature,acid and alkali,salt,antibiotic,and antibacterial ability were tested.The results showed that the strain PA1,SY3-1A,DD21-2 and WM1 had wide adaptability to temperature and environmental pH,which could grow under the conditions of 12~37℃and pH3~9.The salt tolerances of the strains were quite different,9%(PA1),8%(SY3-1A),7%(DD21-2),and 4%(WM1)respectively.The temperature,environmental pH,and salt concentration all had significant effects on the growth of strains(P<0.05).The killer yeasts were more sensitive to antibiotics,having resistance to itraconazole and mycin with low concentration(5μg/mL)or chloroconazole,and different killer yeast strains had different drug resistance characteristics.These four test strains could kill Candida albicans,Metschnikowiabicuspidata and Candida tropicalis,and with strong killing activity,the diameter of inhibition zone of strain PA1、SY3-1A、DD21-2、WM1 against C.albicans YTS-03 reached 23.0,14.1,22.3 and 21.5 mm,respectively.
