摘要
通过70%乙醇提取制得粗黄酮浸膏(黄酮含量为7.75%),且对粗黄酮浸膏进行初步纯化得到精制黄酮浸膏(黄酮含量为20.87%);以L-抗坏血酸棕榈酸酯为阳性对照,比较2种黄酮清除DPPH自由基、超氧阴离子自由基、羟自由基能力。结果表明,当浓度在0.05~0.25 mg/mL时,粗黄酮和精制黄酮对DPPH自由基清除率高达90%左右,而L-抗坏血酸棕榈酸酯清除率最高为77.34%。当浓度在0.40~2.00 mg/mL时,3种样品溶液对超氧阴离子自由基的清除率从大到小依次为精制黄酮、粗黄酮、L-抗坏血酸棕榈酸酯。浓度为2.00 mg/mL时,粗黄酮、精制黄酮和L-抗坏血酸棕榈酸酯对羟自由基的清除率分别为22.41%、36.03%和33.68%。总之,枳椇子黄酮对DPPH自由基、超氧阴离子自由基和羟自由基有较强的清除效果和较好的体外抗氧化活性。
A crude flavonoid extract(flavonoid content of 7.75%)was prepared by 70%ethanol extraction,and crude flavonoid extract preliminary give purified flavonoid extract(flavonoid content of 20.87%);L-ascorbyl palmitate was used as a positive control,the scavenging ability of two flavonoids on DPPH free radicals,superoxide anion free radicals and hydroxyl free radicals was compared.When the concentration within 0.05-0.25 mg/mL,the crude flavones and the purified flavones on DPPH free radical clearance rate was as high as 90%,and L-ascorbic acid palmitate clearance rate was as high as 77.34%.When the concentration within 0.40-2.00 mg/mL,the size of three kinds of sample solution to the superoxide anion free radical clearance rate was purified flavones>crude flavones>L-ascorbic acid palmitate.When the concentration was 2.00 mg/mL,the crude flavones,purified flavones and L-ascorbic acid palmitate to hydroxyl free radical clearance rate was 22.41%,36.03%and 33.68%,respectively.In short,the flavonoids of Hovenia acerba have strong scavenging effects on DPPH free radicals,superoxide anion free radicals and hydroxyl free radicals,and have good antioxidant activity in vitro.
