摘要
目的探讨5-氨基酮戊酸(ALA)光动力疗法(PDT)对耐甲氧西林金黄色葡萄球菌(Methicillin-resistant Staphylococcus aureus,MRSA)的抑制作用。方法选择2017年1月-2018年3月医院住院感染患者血液、鼻咽部、切口、痰液分离的42株MRSA,将不同部位的MRSA分为ALA-PDT组、ALA组、PDT组及对照组,采用标准菌株确定光动力参数。ALA-PDT组给予ALA增敏及PDT光照,ALA组给予ALA干预,PDT组给予PDT光照干预,观察各组及不同来源MRSA菌株活力变化。结果标准菌株在ALA 10mmol/L,PDT 360J/cm^2条件时菌株活力最低,确定其为ALA-PDT光动力干预参数。ALA 10mmol/L,PDT 360J/cm^2条件下,各组内不同来源MRSA菌株活性无显著差异;不同来源的MRSA菌株活性均为ALA-PDT组低于ALA组、PDT组及对照组,差异有统计学意义(P<0.05),ALA组、PDT组及对照组比较差异无统计学意义。结论ALAPDT对MRSA的活性具有抑制作用,并且抑制作用与ALA的浓度、PDT的照射强度有关,并对不同部位分离出的细菌抑制作用没有显著差异。
OBJECTIVE To explore the inhibitory effect of 5-aminolevulinic acid(ALA)photodynamic therapy(PDT)on methicillin-resistant Staphylococcus aureus(MRSA).METHODS A total of 42strains of MRSA were isolated from blood,nasopharynx,incision site and sputum specimens of the patients with infection who were hospitalized from Jan 2017to Mar 2018were divided into the ALA-PDT group,the ALA group,the PDT group and the control group according to the sites where the MRSA strains were isolated.The photodynamic parameters were determined by standard strains.The ALA-PDT group was given ADA sensitization and PDT light,the ALA group was given ALA intervention,and the PDT group was given PDT light intervention,and the changes of activities of the MRSA strains isolated from different specimens sources were observed.RESULTS The activity of the standard strains was the lowest when ALA 10mmol/L and PDT 360J/cm^2 were used,which were determined to be LAL-PDT photodynamic intervention parameters;there was no significant difference in the activity of the MRSA strains among the 4groups under the conditions of ALA 10mmol/L and PDT 360J/cm^2;the activity of the MRSA strains isolated from different specimen sources was significantly lower in the ALA-PDT group than in the ALA group,the PDT group and the control group(P<0.05),while there was no significant difference among the ALA group,the PDT group and the control group.CONCLUSION ALA-PDT can inhibit the activity of MRSA,which is associated with the concentration of ALA and the irradiation intensity of PDT,and there is no significant difference in the inhibitory effect on the strains that are isolated from different sites.
