猪伪狂犬病病毒河北变异株的分离鉴定及其gE基因序列分析 被引量:7

摘要 河北省某猪场猪群已免疫伪狂犬病(pseudorabies,PR)gE基因缺失苗,但仍发生了伪狂犬病疫情。本研究经PCR检测、组织病理学观察、免疫组化染色、病毒分离鉴定、动物试验等,从疑似PR病料中分离鉴定了1株伪狂犬病病毒(pseudorabies virus,PRV),并对该病毒进行gE基因序列分析。结果表明,分离毒gE基因的开放阅读框由1 740 bp组成,编码580个氨基酸。该病毒gE基因与23株PRV参考毒株之间具有较高的同源性,核苷酸和氨基酸的同源性分别为97.5%~99.9%和95.3%~99.7%,与HLJMDJ2013、HeBLP2014、AHSZ-16株核苷酸同源性最高,均为99.9%。与2012年以来分离的变异株(Fa株除外)的gE基因存在相同的独特分子变异特征。遗传进化分析表明,该病毒与其他亚洲毒株同属第1群。动物试验表明该株病毒对兔具有很强的致病性,攻毒组兔全部发生死亡,并能引起兔出现典型的伪狂犬病神经症状。本研究为河北省生猪健康养殖与疾病防疫提供了数据基础和警示预警。 Pseudorabies(PR) broke out on a pig farm in Hebei province, in which the pigs had immunized with the gE gene lacking PR vaccine.To find the causes of the outbreak, a pseudorabies virus(PRV) was isolated in this study, which was named HBXTPRV, from the suspected tissue samples from the diseased pigs, by PCR detection, histopathological observation, immumohistochemical staining, virus isolation and animal test.The gE gene of the strain was sequenced.The results showed that the open reading frame of the gE gene was composed of 1 740 nucleotides, encoding 580 amino acids.The homology analysis of the nucleotide sequences and amino acids of gE indicated that the PRV isolate was in 97.5% to 99.9% and 95.3% to 99.7% homology,respectively, with 23 reference strains, and it was in the highest homology with the strains HLJMDJ2013, HeBLP2014, and AHSZ-16.The gE gene of HBXTPRV possessed the same unique molecular characteristics with all the variant strains isolated since 2012, except the Fa strain.Phylogenetic analysis showed that the isolate was classified as a cluster 1, similar to the other strains isolated in Asia in recent years.The viral strain showed high pathogenicity in rabbits, leading to death of all the infected rabbits manifesting typical PR neurological symptoms.This study provided data base and warning for healthful swine breedingand and control of swine diseases in Hebei Province.
