摘要 利用RAD-seq简化基因组测序鉴定金湖乌凤鸡与其它18个地方鸡种、2个引入肉鸡品种的SNP标记,计算遗传统计量,分析金湖乌凤鸡的遗传多样性和遗传结构,鉴定受选择基因.结果表明,在金湖乌凤鸡中鉴定出SNP标记325 531个,SNP标记在染色体上均匀分布.金湖乌凤鸡的观察杂合度Ho为0.213 6、核苷酸多样度Pi为0.240 4,近交系数Fis为0.111 6,与其它18个地方鸡种相比遗传多样性较为丰富;金湖乌凤鸡的平均遗传分化系数Fst为0.155 6,平均基因流Nm为1.398 2,在系统发育树中形成一个相对独立的分支.通过Fst和θπ检验,在金湖乌凤鸡中发现23个受选择区域,包含104个受选择基因,功能分析表明这些受选择基因主要富集在代谢过程、发育过程、细胞信号转导等生物学通路. RAD-seq simplified genome sequencing technology was applied to identify SNP markers in Jinhu Wufeng chicken, other 18 indigenous breeds, and two introduced ones. Genetic diversity and structure, and selected genes were interpreted based on genetic parameters concerning evolution. A total of 325 531 SNP markers were identified in Jinhu Wufeng chickens, and they were evenly distributed across the chromosomes. The average observed heterozygosity, nucleotide diversity and inbreeding coefficient of Jinhu Wufeng chicken arrived at 0.213 6, 0.240 4 and 0.111 6, respectively, indicating a relative higher genetic diversity compared with other 18 indigenous breeds. The average genetic differentiation coefficient was 0.155 6, with the gene flow averaging at 1.398 2. Jinhu Wufeng chicken stood out as a single branch in the phylogenetic tree constructed using two introduced chicken breeds as outgroups. Based on Fst and θπ testing 23 selected areas and 104 candidate genes were identified. Function analysis further revealed that these selected genes were mainly enriched in the pathways relating to metabolism, development and signal transduction.
