摘要
目的对比长白山地区龙胆、条叶龙胆、三花龙胆质量,对其进行质量评价。方法建立龙胆药材指纹图谱。另建立一标多测方法(QAMS),以龙胆苦苷为参照,计算獐牙菜苦苷、当药苷的相对校正因子,检验方法的可行性并计算含量,对经一标多测方法计算得到的各成分含量与通过外标法测得的实际含量进行对比,并对测定结果进行单因素方差分析。结果建立龙胆药材指纹图谱,共13个共有峰,其中3个峰为龙胆苦苷、獐牙菜苦苷和当药苷,24批龙胆药材相似度均>0.95。2种方法计算得到各成分的含量无显著差异。龙胆、条叶龙胆中龙胆苦苷和当药苷含量高于三花龙胆,獐牙菜苦苷含量无显著差异。结论指纹图谱结合一标多测方法经过实际验证,有较高准确性和可行性,可用于龙胆质量评价。综合评价,龙胆、条叶龙胆质量优于三花龙胆。
Objective To compare and evaluate the quality of Gentiana scabra Bge.,Gentiana manshurica Kitag.and Gentiana triflora Pall.collected in Changbai Mountain Area.Methods The fingerprint of Longdan (Chinese gentian) was established.And quantitative analysis of multi-components by single marker (QAMS) was established,using gentiamarin as the external standard,to calculate the relative correction factor for swertiamarin and sweroside,and test the feasibility and calculate the content,and the content of each component was calculated by QAMS.The actual content measured by the external standard method was compared,and the measurement results were analyzed by one-way ANOVA.Results A total of 13 common peaks were established in the fingerprint of Longdan (Chinese gentian) medicinal materials.Three of them were gentiopicrin,swertiamarin,and sweroside.The similarity of 10 batches of Longdan (Chinese gentian) medicinal materials was all greater than 0.95.There was no significant difference in the content of each component calculated by the two methods.The content of gentiopicrin and sweroside in the medicinal materials obtained from Gentiana scabra Bge.and Gentiana manshurica Kitag.was higher than that obtained from medicinal materials of Gentiana triflora Pall.There was no significant difference in the content of swertiamarin.Conclusion The fingerprint and QAMS have been verified by the actual method,which has high accuracy and feasibility,and can be used for the quality evaluation of gentian.Overall,the quality of Gentiana scabra Bge.and Gentiana manshurica Kitag.is better than Gentiana triflora Pall.
