Hp感染与胃癌组织中PRC2和H3K27me3蛋白的相关性分析 被引量:5

Correlation analysis of PRC2 and H3K27me3 in gastric cancer tissue with Helicobacter pylori infection

摘要 目的探究幽门螺杆菌(Helicobacter pylori,Hp)感染与胃癌组织中多梳抑制复合物2(Polycomb Repressive Complex 2,PRC2)与H3K27me3组蛋白之间的相关性,探究Hp感染在胃癌发病中的作用。方法选取2017年1月-2018年5月于新疆医科大学第三临床医学院接受治疗的106例胃癌患者为研究对象,根据患者是否发生Hp感染将患者分为两组,其中发生Hp感染的49例患者为试验组,未发生Hp感染的57例患者为对照组。使用快速尿素酶检测法、PCR检测以及Giemsa染色法检测患者Hp感染情况;使用免疫组织化学法检测两组患者胃癌组织中PRC2家族成员以及H3K27me3组蛋白的表达情况并进行比较;检测试验组患者胃癌组织与癌旁组织中PRC2家族成员以及H3K27me3组蛋白的表达情况并进行比较;使用Logistic多因素探究Hp感染与胃癌组织中PRC2和H3K27me3组蛋白的相关性。结果快速尿素酶检测法、PCR检测以及Giemsa染色法检测的Hp感染率分别为44.34%、40.57%、46.23%,无统计学差异。试验组患者PRC2阳性率83.67%、H3K27me3组蛋白阳性率79.59%,高于对照组患者的24.56%、19.30%,差异均具有统计学意义(P<0.05)。试验组患者胃癌组织中PRC2阳性率73.46%、H3K27me3组蛋白阳性率67.35%,高于癌旁组织的15.79%、12.28%,差异均具有统计学意义(P<0.05)。Logistic多因素回归分析显示,Hp感染与胃癌组织中PRC2和H3K27me3组蛋白的表达存在相关性,Hp感染与PRC2和H3K27me3组蛋白含量呈正相关关系,差异具有统计学意义(P<0.05)。结论Hp感染与胃癌组织中PRC2和H3K27me3组蛋白的表达呈正相关关系,发生Hp感染的患者PRC2和H3K27me3组蛋白更易表达,在胃癌发病中有一定的作用。 OBJECTIVE To explore the correlation between Helicobacter pylori(Hp)infection and Polycomb repressive complex 2(PRC2)and H3 K27 me3 histone in gastric cancer tissue,and to explore the role of Hp infection in the pathogenesis of gastric cancer.METHODS A total of 106 patients with gastric cancer who were treated in the Third Clinical Medical College of Xinjiang Medical University from Jan.2017 to May 2018 were enrolled.The patients were divided into two groups according to whether they had Hp infection.Among them,49 patients with Hp infection were included in the observation group,and 57 patients who did not develop Hp infection were taken as the control group.Rapid urease test,PCR detection and Giemsa staining were used to detect Hp infection.Immunohistochemistry was used to detect the expression of PRC2 family members and H3 K27 me3 in gastric cancer tissues of the two groups.The expression of PRC2 family members and H3 K27 me3 in gastric cancer tissues and adjacent tissues were compared and the correlation between Hp infection and PRC2 and H3 K27 me3 in gastric cancer tissues was investigated by using Logistic multivariate analysis.RESULTS The infection rates of Hp detected by rapid urease assay,PCR and Giemsa staining were 44.34%,40.57%and 46.23%,respectively,with no significant difference.The positive rates of PRC2 and H3 K27 me3 in the experimental group were 83.67%and 79.59%respectively,which were significantly higher than those in the control group(24.56%and 19.30%,respectively)(P<0.05).The positive rates of PRC2 and H3 K27 me3 in gastric cancer tissues of the experimental group were 73.46%and 67.35%,respectively,significantly higher than those in adjacent tissues(15.79%and 12.28%,P<0.05).Logistic multivariate regression analysis showed that Hp infection was significantly correlated with the expression of PRC2 and H3 K27 me3 in gastric cancer tissues.Hp infection was positively correlated with the content of PRC2 and H3 K27 me3(P<0.05).CONCLUSION Hp infection has a positive correlation with the expression of PRC2 and H3 K27 me3 in gastric cancer tissues.PRC2 and H3 K27 me3 are more easily expressed in patients with Hp infection,and have a certain role in the pathogenesis of gastric cancer.
