紫花苜蓿(Medicago sativa)YABBY基因家族的生物信息学分析及其对逆境胁迫的响应 被引量:12

摘要 YABBY转录因子家族是植物特有转录因子,与植物形态有关,在植物响应胁迫方面也有重要的调控作用。为了研究YABBY基因家族在苜蓿抗逆性中的作用,本研究利用生物信息学对苜蓿的YABBY转录因子家族成员、分布及结构等进行分析。对苜蓿、大豆、水稻和拟南芥进行系统进化树的分析,表明该家族基因在植物中具有同源性。理化性质和结构分析表明,苜蓿中YABBY基因主要分布在第2、4、5条染色体上,苜蓿YABBY家族基因中包含5个保守结构域,其蛋白质主要以无规则卷曲的形式构成,α-螺旋、β-折叠和β-转角含量较少,启动子序列分析发现该家族存在ABRE顺式反应元件,能够响应盐胁迫及ABA(Ascisic acid)胁迫,通过基因表达模式分析,发现MsYABBY2基因响应盐、碱、干旱等逆境胁迫,推测其在抗逆境胁迫中起到关键调节作用。本研究为后续研究MsYABBY基因的抗逆机制及调控机理提供了理论基础。 YABBY transcription factor family is a plant-specific transcription factor, which is related to plant morphology and plays an important regulatory role in plant response to stress. In order to study the role of YABBY gene family in the resistance of alfalfa, this study used bioinformatics to analyze the members, distribution and structure of YABBY transcription factor family in alfalfa genome. Analysis of phylogenetic trees in alfalfa,soybean, rice and Arabidopsis showed that the family genes are homologous in plants. The physicochemical properties and structural analysis showed that the YABBY gene was mainly distributed on chromosomes 2, 4 and5, and the YABBY family contained five conserved domains. The protein was mainly composed of random coil,α-helix, β-sheet and β-turn content is less. Promoter sequence analysis revealed that the family had ABRE cis-responsive elements that responded to salt stress and ABA stress. Through gene expression pattern analysis, we found that Ms YABBY2 gene responded to stresses such as salt, alkali, drought and so on. It is supposed that MsYABBY2 gene plays a key role in resistance to stress. This study provides a theoretical basis for the further study of the stress resistance mechanism and regulation mechanism of MsYABBY gene.
