摘要
目的:建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)测定茯苓水溶性多糖中单糖含量的测定方法。方法:采用Shodex Asahipak NH2P-504E色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-水(81∶19);流速:1.0 ml·min^-1;柱温:35℃;进样量:50μl;ELSD参数:漂移管温度:70℃,喷雾管温度:30℃,载气流速:1.1 ml·min^-1。结果:岩藻糖、甘露糖、半乳糖、葡萄糖的进样量分别在1.18~3.68μg(r=0.9987)、4.26~13.33μg(r=0.9973)、7.09~22.18μg(r=0.9963)、8.43~26.35μg(r=0.9970)呈良好线性关系;平均加样回收率分别为100.64%,104.01%,106.10%,105.74%,RSD分别为3.70%,2.09%,1.78%,1.74%(n=6)。结论:该方法准确可靠、操作简便、分离效果好,适用于茯苓水溶性多糖中单糖的含量测定。
Objective:To establish an HPLC-ELSD method for the determination of monosaccharides in Poria cocos water-soluble polysaccharides.Methods:A Shodex Asahipak NH2P-504E column(250 mm×4.6 mm,5μm)was used,the mobile phase was acetonitrile-water(81∶19),the flow rate was 1.0 ml·min^-1,the column temperature was 35℃,and the injection volume was 50μl.The parameters of the accelerator were as follows:the drift tube temperature was 70℃,the spray tube temperature was 30℃,the carrier gas flow rate was 1.1 ml·min^-1.Results:The injection amount of fucose,mannose,galactose and glucose was 1.18-3.68μg(r=0.9987),4.26-13.33μg(r=0.9973),7.09-22.18μg(r=0.9963)and 8.43-26.35μg(r=0.9970),respectively,all showing a good linear relationship.The average recovery of sample addition was 100.64%,104.01%,106.10%and 105.74%,and the RSD was3.70%,2.09%,1.78%and 1.74%(n=6),respectively.Conclusion:The method is accurate,reliable,simple and effective in the determination of monosaccharides in Poria cocos water-soluble polysaccharides.
