摘要
目的:探究补肾活血汤通过抑制Wntβ-catenin信号通路对激素性股骨头坏死大鼠疗效及血管形态、血管内皮生长因子受体(VEGF)的影响。方法:SD大鼠随机分为假手术(sham)组、模型组和干预组。通过臀肌下注射醋酸泼尼松龙建立模型组模型。干预组在建模后使用补肾活血汤灌胃。HE染色被用于检测股骨头组织学情况。CT扫描股骨头血管情况。Western Blot检测股骨头组织中Wnt、β-catenin、VEGF蛋白水平。结果:建模后骨小梁稀疏变细,并出现明显的断裂和缺损,骨细胞数目少;干预组骨小梁缺损显著轻于模型组。模型组骨小梁面积显著降低而空骨陷窝率显著增加(P<0.01),干预组的骨小梁面积显著高于模型组而空骨陷窝率显著低于模型组(P<0.01)。Sham组大鼠股骨头血管呈现网状,分布较为均匀,结构清晰;模型组大鼠股骨头内血管明显减少,微结构不清晰;干预组股骨头内血管呈现网状,部分呈网状。模型组血管体积分数和血管厚度显著低于sham组(P<0.01),干预组血管体积分数和血管厚度显著高于模型组(P<0.01)。模型组的Wnt、β-catenin、VEGF蛋白水平显著低于Sham组(P<0.01),干预组的Wnt、β-catenin、VEGF蛋白水平显著高于模型组(P<0.01)。结论:补肾活血汤具有上调Wntβ-catenin信号通路以及VEGF表达的作用,进而促进骨骼组织血管的生成,治疗模型组。
Objective:To explore the effect of Bushen Huoxue Decoction on the therapeutic effect,vascular morphology and VEGF of rats with steroid-induced femoral head necrosis by inhibiting Wntp-catenin signaling pathway.Methods:SD rats were randomly divided into sham group,model group and intervention group.The model group was established by intraperitoneal injection of prednisolone acetate.The intervention group was administered with Bushen Huoxue Decoction after modeling.HE staining was used to detect the histology of the femoral head.CT scan of the femoral head vascular condition.Western Blot was used to detect the levels of Wnt,p-catenin and VEGF protein in the femoral head tissue.Results;After modeling,the trabecular bones were sparse and thin,and there were obvious fractures and defects,and the number of bone cells was small.The trabecular bone defect in the intervention group was significantly lighter than that in the model group.The area of trabecular bone in the model group group was significantly decreased and the rate of empty lacunae was significantly increased(P<0.01).The trabecular bone area of the intervention group was significantly higher than that of the model group and the empty bone lacuna rate was significantly lower than that of the model group(P<0.01).In the Sham group,the femoral head vessels were reticular,distributed evenly and clearly structured.In the model group,the blood vessels in the femoral head were significantly reduced,and the microstructure was not clear.The blood vessels in the femoral head of the intervention group were reticulated and partially reticular.The volume fraction of blood vessels and blood vessel thickness in the model group were significantly lower than those in the sham group(P<0.01).The volume fraction of blood vessels and blood vessel thickness in the intervention group were significantly higher than those in the model group(P<0.01).The levels of Wnt,p-catenin and VEGF in model group were significantly lower than those in the sham group(P<0.01).The levels of Wnt,p-catenin and VEGF protein in the intervention group were significantly higher than those in the model group(P<0.01).Conclusion:Bushen Huoxue Decoction can up-regulate the Wntp-catenin signaling pathway and the expression of VEGF,thereby promoting the formation of vascular tissue and treating model.
