摘要
目的:通过观察健肝消脂方对非酒精性脂肪性肝病(NAFLD)动物模型活性氧簇(ROS)水平的影响,探讨健肝消脂方干预胰岛素抵抗(IR)进而治疗NAFLD的分子机制。方法:采用高糖高脂饲料喂养大鼠复制NAFLD动物模型。将NAFLD模型大鼠随机分为正常对照组、病理模型组、二甲双胍组和健肝消脂方低剂量组、中剂量组、高剂量组,每组15只。药物干预后,检测各组大鼠空腹胰岛素水平、空腹血糖水平、肝脏ROS水平,计算胰岛素抵抗指数(HOMA-IR)、分析肝脏脂质沉积情况。结果:与正常对照组比较,病理模型组大鼠空腹胰岛素含量和HOMA-IR指数、ROS水平明显升高(P<0.05),肝脏脂肪变性明显增强(P<0.05)。健肝消脂方高剂量组和二甲双胍组能抑制ROS,改善大鼠胰岛素抵抗(P<0.05),并改善肝脏脂质沉积(P<0.05)。结论:健肝消脂方对NAFLD疗效确切,其作用机制可能与其抑制ROS活性进而改善胰岛素抵抗有关。
Objective:To further elucidate the molecular mechanism of intervention of Jiangan Xiaozhi Recipe for non-alcoholic fatty liver disease(NAFLD)by relieving insulin resistance(IR)which was closely related to the ROS activity of animal model.Methods:Rats were fed with high-sugar and high-fat diet for the reproduction of a ratmodel of NAFLD.Then NAFLD model rats were randomly divided into normal controlled group,pathological model group,metformin group,Jiangan Xiaozhi Recipe low—dose group,middle dose group,high dose group,15in each group.Fasting insulin levels,fasting plasma glucose levels and liver reactive oxygen species(ROS)levels,were measured after drug intervention,and insulin resistance index(HOMAIR)and liver lipid deposition were calculated.Results:Compared with normal controlled group,fasting insulin content,HOMAIR index and ROS level were significantly increased in the pathological model group(P<0.05),and liver steatosis was significantly enhanced(P<0.05).Jiangan Xiaozhi Recipe high-dose group and metformin groupcould inhibit ROS and relieve insulin resistance and reduce liver steatosisin rats(P<0.05).Conclusion:Jiangan Xiaozhi Recipe is effective for NAFLD,and its mechanism may be related to the alleviation of insulin resistance by theinhibition of ROS.
