尖锐湿疣疣体内TLR9表达水平与免疫应答和细胞凋亡的相关性研究 被引量:14

摘要 目的研究尖锐湿疣(CA)疣体内Toll样受体9(TLR9)表达水平与免疫应答和细胞凋亡的相关性。方法选择2014年8月至2018年7月期间在葫芦岛市中心医院诊断为初发CA和复发CA的患者、分别作为初发组和复发组,另取同期接受包皮环切术的患者作为对照组。收集CA病灶和正常包皮组织,测定TLR9、Th1/Th2/Th17/Treg转录因子、凋亡基因的mRNA表达水平及Th1/Th2/Th17/Treg细胞因子的含量。结果初发组、复发组CA病灶内TLR9、T-bet、RORγt、增殖细胞核抗原(PCNA)、Survivin的mRNA表达水平及干扰素-γ(IFN-γ)、白细胞介素(IL)-17的含量明显高于对照组,GATA-3、FOXP3、Fas、FasL、Caspase-3的mRNA表达水平及IL-4、IL-10的含量明显低于对照组;且复发组CA病灶内TLR9、T-bet、RORγt、Fas、FasL、Caspase-3的mRNA表达水平及IFN-γ、IL-17的含量明显低于初发组,GATA-3、FOXP3、PCNA、Survivin的mRNA表达水平及IL-4、IL-10的含量明显高于初发组。CA病灶内TLR9的mRNA表达水平与IFN-γ、IL-17的含量及T-bet、RORγt、Fas、FasL、Caspase-3的mRNA表达水平呈正相关,与IL-4、IL-10的含量及GATA-3、FOXP3、PCNA、Survivin的mRNA表达水平呈负相关。结论 TLR9表达水平的改变与尖锐湿疣的发生、复发及免疫应答紊乱、细胞凋亡异常有关。 Objective To study the correlation between Toll-like receptor 9(TLR9) expression with immune response,apoptosis in condyloma acuminatum(CA).Methods Patients diagnosed as primary CA and recurrent CA in our hospital from August 2014 to July 2018 were selected as primary group and recurrent group,and patients undergoing circumcision at the same time were selected as control group.The expression of TLR9,Thl/Th2/Th17/Treg transcription factor,apoptotic gene and the contents of Thl/Th2/Th17/Treg cytokines were measured in CA lesions and normal prepuce tissues.Results The mRNA expression levels of TLR9,T-bet,RORγt,PCNA,Survivin and the contents of IFN-γ,IL-17 in CA lesions of the primary and recurrent groups were significantly higher than those of the control group,the mRNA expression levels of GATA-3,FOXP3,Fas,FasL,Caspase-3 and the contents of IL-4,IL-10 in CA lesions were significantly lower than those of the control group,and the mRNA expression levels of TLR9,T-bet,ROR-γt,Fas,FasL,Caspase-3 and the contents of IFN-γ,IL-17 in CA lesions ofrecurrent group were significantly lower than those of primary group,the mRNA expression levels of GATA-3,FOXP3,PCNA,Survivin and the contents of IL-4,IL-10 in CA lesions were significantly higher than those of primary group.The expression levels of TLR9 in CA lesions were positively correlated with the contents of IFN-γ,IL-17 and the expression levels of T-bet,RORγt,Fas,FasL,Caspase-3 and negatively correlated with the contents of IL-4,IL-10 and the expression levels of GATA-3,FOXP3 and PCNA,Survivin.Conclusion The change of TLR9 expression level is related to the occurrence,recurrence,immune response disorder and abnormal apoptosis of condyloma acuminatum.
