摘要
目的探究右美托咪定(dexmedetomidine,DEX)对骨巨细胞瘤细胞增殖、侵袭及迁移的影响.方法将细胞分Control、DEX 0.01、0.1及1μmol/L组.BrdU检测细胞增殖,transwell检测细胞侵袭,划痕实验检测细胞迁移,免疫印迹检测Ki67、PCNA、Bcl-2、VEGF、MMP-2、MMP-9、NF-κB p65及其下游蛋白表达水平、p38 MAPK表达及其下游蛋白磷酸化比值.结果与Control组比较,DEX 0.1、1μmol/L组BrdU阳性细胞百分比、侵袭细胞数、划痕闭合率和Ki67、PCNA、Bcl-2、VEGF、MMP-2、MMP-9表达水平显著降低,抑制p38 MAPK、NF-κB信号通路的激活.结论DEX通过调控p38 MAPK及NF-κB表达抑制骨巨细胞瘤细胞增殖、侵袭及迁移.
Objective To investigate the effects of dexmedetomidine on proliferation,invasion and migration of bone giant cell tumor cells.Methods The cells were divided into control,DEX 0.01,0.1 group and1μmol/Lgroup.Cell proliferation was detected by BrdU;cell invasion by tr-answell,cell migration by scratching experiment,and the expression of Ki67,PCNA,Bcl-2,VEGF,MMP-2,MMP-9,NF-κB p65 and their downstream proteins p38 MAPK and its down-stream protein phosphorylation ratio by Western blotting.Results Compared with the control group,the percentage of positive BrdU cells,the number of invasive cells,the scratch closure rate and the expression levels of Ki67,PCNA,Bcl-2,VEGF,MMP-2 and MMP-9 in DEX 0.1 and 1μmol/L groups were significantly decreased,which inhibited the activation of p38 MAPK and NF-κB p65 signaling pathways.Conclusion DEX inhibits the proliferation,invasion and migration of giant cell tumor cells by regulating the expression of p38 MAPK and NF-κB p65.
