猪源肠外致病性大肠杆菌的分离鉴定及致病性 被引量:7

摘要 【目的】从临床病死仔猪体内分离肠外致病性大肠杆菌,研究猪群中流行菌株的毒力基因分布、耐药性、致病性情况.【方法】病死仔猪肠外组织分离大肠杆菌,然后进行16S rDNA PCR测序鉴定、种系发育分群、毒力基因检测、抗菌药物的药敏试验、小鼠致病性试验、全基因组测序验证.【结果】共筛选到30株猪源肠外致病性大肠杆菌,其中A群4株、B1群15株、B2群5株、D群6株;毒力基因检测结果发现vaT、iutA、kpsMII、hlyD基因检出率分别为70.0%、33.33%、23.33%、20.0%;药敏试验结果显示:菌株都对多粘菌素B较敏感,对氨苄西林、林可霉素、杆菌肽以及利福平的耐药率为100%;获取了Ex-P27菌株基因组序列图谱,构建了该菌株基因组圈图.【结论】肠外致病性大肠杆菌分离株主要分布在B1群,对氨苄西林、林可霉素、杆菌肽以及利福平均耐药,为指导猪源肠外致病性大肠杆菌的防治与临床用药提供依据. 【Objective】To study the virulence genes and virulence genes of pig-derived enteropathogenic Escherichia coli,bacteria were isolated from dead piglets.【Method】E.coli in the extraintestinal tissues of sick piglets was isolated and purified,and then tested on 16S rDNA PCR sequencing,germline developmental clustering,virulence gene,antimicrobial drug susceptibility,mouse pathogenicity,whole genome sequencing verification.【Results】A total of 30 pig-derived enteropathogenic E.coli were screened,including 4 strains of A,15 strains of B1,5 strains of B2,and 6 strains of D.The virulence gene detection results showed vaT,iutA,kpsMII,hlyD genes were 70%,33.33%,23.33%,20%respectively.The drug susceptibility test showed that all strains were sensitive to polymyxin B,to ampicillin and lincomycin.The resistance rates of ubiquitin,bacitracin and rifampicin were 100%.The genome sequence map of Ex-P27 strain was obtained,and the genome map of the strain was constructed.【Conclusion】The pig-derived extraintestinal E.coli were enriched in B1 group and were sensitive to polymyxin B,to ampicillin and lincomycin.This study provides the basis for the prevention and treatment of extraintestinal pathogenic E.coli.
