摄食不同种类饵料的鲤肝脏组织miRNA表达谱分析及验证 被引量:1

Analysis and validation of miRNA expression profiles in liver of Cyprinus carpio feeding with different types of diets

摘要 MicroRNA(miRNAs)是一类长度为18-25nt的内源性非编码单链小RNA,参与细胞增殖与凋亡、免疫、脂肪代谢等过程。本试验以三组投喂不同类型饵料的鲤(Cyprinus carpio)肝脏为研究材料,通过HiSeq 2500深度测序技术和生物信息学分析miRNA,鉴定出235个已知miRNAs。三种饵料组共表达的差异miRNAs有13个,随机挑选5个miRNA并采用qPCR进行验证,表达趋势在qRT-PCR结果和RNA测序结果中高度相似;对13个共表达的差异表达miRNAs进行靶基因预测,共预测到靶基因530个。对预测靶基因进行KEGG通路分析,结果显示参与2-羰基羧酸代谢,糖基磷脂酰肌醇(GPI)-锚定生物合成,柠檬酸循环(TCA循环)等通路的调节。本研究首次了解鲤摄食不同种类饵料的miRNAs表达特征,可以为深入了解miRNA对鲤摄食不同种类饵料过程的调控作用奠定基础。 MicroRNAs(miRNAs)are endogenous,non-coding single-stranded small RNAs with the length 18-25nt,which participate in cell proliferation and apoptosis,immunity,lipid metabolism and so on.Adult Cyprinus carpio are often cultured in paddy fields to improve water quality and achieve higher economic benefits in Southwest China.The liver of common carp after 56-days feeding period with three types of feed(animal diet,plant diet and mix diets)was sampled to analyze miRNA transcriptions by Illumina HiSeq 2500 deep sequencing technology and bioinformatics in this study.A total of 235 known miRNAs were identified.There were 13 co-differentially expressed microRNAs in three groups.Five microRNAs were randomly selected and verified by qPCR.The results showed that the expression trend was basically consistent with the sequencing results.The target genes of 13 co-differentially expressed microRNAs were predicted,a total of 530 target genes were predicted.KEGG pathway analysis of predicted target genes showed that differential miRNAs were involved in the regulation of 2-Oxocarboxylic acid metabolism,Glycosylphosphatidylinositol(GPI)-anchor biosynthesis,citrate cycle(TCA cycle)and so on.This study provides a molecular basis for the utilization of different type of diets in fish.
